蛋白磷酸酶2A癌性抑制因子在喉鳞状细胞癌中的表达及临床意义

目的研究蛋白磷酸酶2A癌性抑制因子（cancerous inhibitor of protein phosphatase 2A,CIP2A）在喉鳞状细胞癌（简称喉鳞癌）中的表达及其与喉鳞癌临床病理特征的关系。方法应用实时荧光定量PCR及免疫组化方法检测40例喉鳞癌组织与25例癌旁组织中CIP2A的表达,并分析其与临床病理参数的关系。结果与癌旁组织相比,CIP2A mRNA在喉鳞癌组织的表达量增加（Z=-3.182,P〈0.05）。CIP2A蛋白阳性表达定位于细胞膜与细胞质,癌组织与癌旁组织的阳性表达率分别为70.0%（28/40）和40.0%（10/25）,差异具有统计学意义（χ^2=5.70,P〈0.05）;CIP2A mRNA的表达水平和蛋白阳性表达率在III期和IV期喉癌组明显高于I期和II期病例组（Z=-2.610,χ2=5.560,P〈0.05）,有淋巴结转移组明显高于无转移组（Z=-3.346,χ^2=3.815,P〈0.05）。结论 CIP2A与喉鳞癌的发生及浸润转移密切相关,有望成为喉鳞癌治疗的靶点。

Expression and its clinical significance of CIP2A in laryngeal squamous cell carcinoma

OBJECTIVE To investigate the expression of cancerous inhibitor of protein phosphatase 2A（CIP2A）in laryngeal squamous cell carcinoma（LSCC）and to analyze its relation with the clinicopathologic characteristics of the carcinoma.METHODS CIP2A expression（mRNA and protein）was measured in laryngeal carcinoma tissues（n=40）and para-carcinomatissues（ n = 2 5）using real time PCR and immunohistochemistry（IHC）.The CIP2A expression level was compared with several clinicopathological parameters.RESULTS Expression of CIP2A mRNA was increased in LSCC as compared in the para-carcinoma tissues（Z =-3.182,P〈0.05）.The positive expression of CIP2A protein was located in the cell membrane and cytoplasm.The positive expression rate of CIP2A protein in carcinoma group and para-carcinoma group were 70.0%（28/40）and 40.0%（10/25）respectively,which was statistically signifcant（χ^2=5.70,P〈0.05）.The expression level of CIP2A mRNA and the positive expression rate of protein in laryngeal carcinoma tissues of the advanced stage group（stage III and stage IV）were signifcantly higher than those in laryngeal carcinoma tissues of stage I and II group（Z=-2.610,χ^2=5.560,P0.05）,and they were also significantly higher in laryngeal carcinoma tissues of the patients with lymph node metastasis than those in patients without lymph node metastasis（Z =-3.346,χ^2=3.815,P〈0.05）.CONCLUSION CIP2A may play a significant role in carcinogenesis and metastasis of LSCC and therefore,it may be a potential target for chemotherapy of LSCC.
