木犀草素对糖尿病脑梗死大鼠脑组织HSP70及Fas的表达的影响

摘    要:目的观察木犀草素对糖尿病脑梗死大鼠脑组织中HSP70及Fas的表达的影响。方法60例Wistar大鼠随机分为5组:对照组、模型组及木犀草素高、中、低剂量组。采用高脂高糖饮食加腹腔注射小剂量链脲霉素建立糖尿病大鼠模型,线栓法建立大鼠大脑中动脉永久缺血模型,免疫组化法检测脑组织中HSP70及Fas蛋白的表达。结果低、中、高剂量的木犀草素均能改善糖尿病脑梗死大鼠的神经行为学评分,其中中剂量组及高剂量组效果更明显。HSP70在对照组无表达,模型组表达与对照组比较差异有统计学意义(P〈0.01);低、中、高3个剂量的木犀草素组HSPT0表达均高于模型组(P〈0.01)。模型组大鼠脑组织Fas阳性细胞数显著高于对照组(P〈0.01);木犀草素高、中、低剂量组大鼠脑组织Fas阳性细胞数显著低于模型组(P〈0.01)。结论木犀草素对糖尿病脑梗死大鼠的神经损伤有保护作用,其机制可能与木犀草素升高大鼠脑组织HSP70表达并降低Fas表达,从而减少神经细胞的凋亡有关。

Abstract:Objective To investigate the effect of Luteolin on expression of HSPT0 and Fas in diabetic cerebral infarction of rats brain tissue. Methods Sixty Wistar rats were randomly divided into 5 groups : normal control, model, high - dose Luteolin, mid - dose Luteolin and low - dose Luteolin group. Rat models of diabetes mellitus were built by high sucrose, fat diet and small dose of streptozotocin injection. Models of rats permanent middle cerebral artery occlusion were built by the suture method Immunohistochemistry staining was used to detect the of HSP70 and Fas protein expressions in brain tissue of rats. Results Low, medium and high dose of Luteolin can all improve rats' neurobehavioral scores of diabetes cerebral infarction, the effect of medium and high dose group are more obvious. There was no expression of HSPT0 in the control group, the difference in model compared with control group was significant (P 〈0.01 ) ; the level of HSP70 expression in low, medium and high dose of Luteolin groups all higher than the compared group (P 〈 0. 01 ) . The amount of Fas positive ceils was obviously higher in model group than that of control group (P 〈 O. O1 ) ; In low, medium and high dose of Luteolin group the amount of Fas positive cells was obviously lower than that of model group ( P 〈 O. 01 ) . Conclusions Luteolin plays an important role in protecting against neurological impairment of diabetes complicating with focal cerebral infarction rats, its mechanism may be related to increasing the expression of HSP70 and decreasing the expression of Fas, thereby reducing the apoptosis of nerve cells.
