附子生物碱对人胃癌细胞株SGC-7901 凋亡及细胞周期的影响

摘    要:目的:观察附子生物碱对人胃癌细胞株SGC-7901 的增殖以及凋亡的影响。方法:运用MTT 法,测定经附子生物碱处理后体外培养SGC-7901 细胞的增殖活性。分别采用Annexin-V/PI 双染法、流式细胞仪检测附子生物碱对细胞凋亡周期的影响。结果:淤附子生物碱分别作用人胃癌细胞株SGC-7901 24 h、48 h、72 h 后的IC50 为0.231 8±0.002 2、0.160 1±0.022 7、0.103 1±0.023 1 mg·mL-1,与空白对照组比较有显著性差异(P<0.01)。于当附子生物碱浓度达到0.8 mg·mL-1 时出现明显的凋亡,凋亡率为(59.38±5.05)%。盂流式细胞仪检测发现,与空白对照组相比,给药组处于S 期细胞的比例显著增多,并且G0/G1 期前出现凋亡亚二倍体峰。结论:附子生物碱体外对人胃癌细胞SGC-7901 具有增殖抑制作用,并促进其凋亡,还可使SGC-7901 细胞阻滞于S 期。

Abstract:This study was aimed to investigate effect of aconite alkaloids on proliferation and apoptosis of human gastric cancer cell line SGC-7901. Effects of different concentrations of aconite alkaloids on proliferation of human gastric cancer cell line SGC-7901 were investigated with MTT assay; induced apoptosis and cell cycle blocking were detected with flow cytometer (FCM). The results showed that the IC50 of aconite alkaloids on human gastric cancer cell line SGC-7901 was 0.2318 ±0.0022, 0.1601 ±0.0227, 0.1031 ±0.0231 mg/ml at 24 h, 48 h and 72 h, respectively, compared to the control group with significant difference (P <0.01). When the aconite alkaloids concentration was 0.8 mg/ml, it appeared with obvious apoptosis. The apoptosis rate was (59.38 ±5.05)%. The FCM detection showed that compared with control group, the percentage of S-phase cells increased in the treatment group. And a typical sub-diploid peak appeared before G0 / G1 phase. It was concluded that aconite alkaloids can inhibit the proliferation of human gastric cancer cell line SGC-7901 in vitro, induce the apoptosis of cells and make SGC-7901 cells remain in S phase.
