紫杉醇联合肿瘤坏死因子相关凋亡诱导配体对前列腺癌细胞的抑制作用

摘    要:目的 探讨紫杉醇联合肿瘤坏死因子相关凋亡诱导配体(TRAIL)对前列腺癌细胞的抑制作用。方法 MTT法测定紫杉醇、TRAIL及紫杉醇联合TRAIL作用于LNCAP、Du145、PC3细胞后的细胞生存率,同时,用流式细胞技术测定3种细胞的凋亡率。结果 2ng/ml TRAIL对LNCAP、PC3细胞有明显的抑制作用,但对Du145细胞抑制作用不明显;当TRAIL与紫杉醇联用则对3种癌细胞均显示出明显的抑制作用,且发生在给药24h后,72h时抑制作用最强。TRAIL对3种癌细胞有诱导凋亡作用,5ng/ml较2ng/ml作用强;紫杉醇与TRAIL联用明显增强诱导凋亡作用。结论 紫杉醇与TRAIL联合应用能明显增强对LNCAP、Du145、PC3细胞的抑瘤效果和诱导凋亡作用。

Abstract:Objective To investigate the inhibitory effect of paclitaxe in combination with TNF-related apoptosis-inducing ligand(TRAIL) on the prostate cancer cells.Methods The survival rate of LNCAP,Du145 and PC3 cells treated with paclitaxe,TRAIL or paclitaxe-TRAIL was tested by MTT methods.The apoptosis rate of LNCAP,Du145 and PC3 cells was detected by flow cytometry.Results Two ng/ml TRAIL could obviously inhibit LNCAP and PC3 cells,but had no significant suppression on Du145 cells.The combined use of TRAIL and paclitaxe displayed significantly inhibitory effects on the above 3 cell lines in 24 h,strongest in 72 h.TRAIL could induce apoptosis of the above 3 cell lines,stronger in 5 ng/ml than in 2 ng/ml.The combined use of paclitaxe-TRAIL could obviously enhance apoptosis-inducing effects.Conclusion Combined use of TRAIL and paclitaxe could obviously enhance the inhibitory effects and apoptosis-inducing effects on LNCAP,Du145 and PC3 cells.
