趋化因子CXCL5基因多态性与2型糖尿病胰岛素抵抗相关性研究

摘    要:目的研究2型糖尿病患者趋化因子CXCL 5基因启动子-156位点G/C多态性分布,探讨该基因多态性与胰岛素抵抗的关系。方法采用多聚酶链式反应限制性片段长度多态性技术检测210例2型糖尿病患者及171例非糖尿病对照者趋化因子CXCL5基因启动子-156位点G/C多态性。测量身高、体重、并计算体重指数(BMI)、检测血脂、放射免疫分析法测空腹胰岛素,计算胰岛素抵抗指数(HOMA-IR)。结果 2型糖尿病组空腹血糖、甘油三酯、总胆固醇、低密度脂蛋白、HOMA-IR高于对照组(P<0.001),2型糖尿病组高密度脂蛋白低于对照组(P<0.05)。2型糖尿病组中携带C基因型(G/C+C/C)的患者与携带G/G基因型的患者年龄、性别、甘油三酯、高密度脂蛋白差异无统计学意义(P>0.05)。携带C基因型(G/C+C/C)的患者空腹血糖、空腹胰岛素、总胆固醇、低密度脂蛋白、HOMA-IR、BMI均高于携带G/G基因型的患者(P<0.05)。结论CXCL5基因启动子-156位点G/C多态性与2型糖尿病发病无关,2型糖尿病患者CXCL5变异基因携带者有较高的HOMA-IR与BMI,表明CXCL5基因突变可能与胰岛素抵抗有关。

Abstract:【Objective】 To investigate the distribution of CXCL5 promoter gene-156G/C polymorphisms in patients with type 2 diabetes mellitus(T2DM) and to analyze the association between CXCL5 gene-156G/C polymorphism and insulin resistance.【Methods】 The polymorphism of CXCL5 gene promoter region-156G/C was detected by PCR-RFLP method in 210 patients with T2DM and 171 controls.All patients’ height and weight were measured,and their BMI were calculated.Moreover,their TC,TG,LDL-C,HDL-C,FBG,FINS were measured with automatic biochemical analyzer and their HOMA-IR were then calculated.【Results】 FBG,TG,TC,LDL-C,HOMA-IR of T2DM group are statistically significantly higher than the control group(P <0.001).HDL-C of T2DM group is statistically significantly lower than the control group(P <0.05).In T2DM group,comparison of C genotype(G/C+C/C) carriers and G/G genotype carriers show no statistically significant difference in terms of age,sex,TG,and HDL(P >0.05).FBG,FIN,HOMAIR,BMI,TC and LDL for C genotype(G/C+C/C) carriers are statistically significantly higher than for G/G genotype carriers(P <0.05).【Conclusion】 The distribution of CXCL5 promoter gene-156G/C polymorphism has no relation with T2DM.In T2DM group and the control group,CXCL5 mutant gene carriers are found to have considerably high HOMAIR and BMI.Therefore,CXCL5 gene mutation is related to insulin resistance.
