荧光探针PCR检测金黄色葡萄球菌的研究

目的 研究并建立荧光探针聚合酶链反应(PCR)技术,用于金黄色葡萄球菌(SAU)及耐甲氧西林金黄色葡萄球菌(MRSA)的临床检测.方法 采用标准化荧光探针PCR技术及操作流程,进行灵敏性及特异性的鉴定.结果 分析性灵敏性达到1×10 2拷贝/reaction,达到设计要求,具备高重复性；对1196例临床标本应用荧光探针PCR检测SAU,临床灵敏性为97.41％,临床特异性为98.47％,对527例SAU的临床样本进行MRSA筛查,临床灵敏性为99.74％,临床特异性为95.59％；对385份鼻腔拭子标本应用荧光探针PCR检测SAU,检出率达18.18％；对其中40株SAU进行MRSA筛查,检出率达42.50％.结论 荧光探针PCR技术检测SAU及其MRSA的方法具有较高的优越性,适合在临床检测应用.

Study on detection of Staphylococcus aureusby fluorescence PCR

OBJECTIVE To establish and study Fluorescence PCR method for clinical detection of Staphylococcus aureus(SAU) and methicillin-resistant Staphylococcus aureus(MRSAs).METHODS The drug susceptibility and specificity was performed by using standard fluorescence PCR method and procedure.RESULTS The highest value of the analytic sensitivity was 1×102 copies/reaction,meeting the requirements of the design,and the method was highly reproducible;the clinical sensitivity and specificity of detection of S.aureus from 1196 specimens by fluorescence PCR were 97.41% and 98.47%,respectively;the clinical sensitivity of detection of MASAU from 527 S.aureus samples was 99.74%,and the specificity was 95.59%;the detection rate of S.aureus from 385 nasal swab samples was 18.18%,and the detection rate of MRSA was 42.50% in these 40 S.aureus samples.CONCLUSION Fluorescence PCR has great advantages in clinical detection of S.aureus and MRSA.
