阿托伐他汀对急性心肌梗死髓过氧化物酶的影响

摘    要:目的探讨阿托伐他汀对急性心肌梗死髓过氧化物酶的影响。方法雄性SD大鼠结扎左冠状动脉前降支建立心肌梗死模型,并随机分为治疗组(19只)、对照组(19只)和假手术组(18只),治疗组术前给予阿托伐他汀10mg.kg-1.d-1,对照组和假手术组给予相应体积的生理盐水。瑞吉染色法检测血液中中性粒细胞(polymorph nuclear neutrophil,PMN)的数量;HE染色光镜检测心肌组织中PMN浸润的数量;TTC染色法测定心肌梗死范围;髓过氧化物酶(MPO)法测定血清和心肌MPO活性。结果治疗组和对照组血液、心肌内的PMN浸润较假手术组明显增加(P<0.01),但治疗组较对照组PMN数量明显减少(P<0.05)。治疗组和对照组血清、心肌内MPO活性均较假手术组明显增加(P<0.01);与对照组比较,治疗组血清、心肌内MPO活性明显降低(P<0.05)。治疗组和对照组心肌梗死范围与假手术组比较有显著差异(P<0.01);治疗组与对照组比较,心肌梗死范围明显缩小(P<0.05)。结论急性心肌梗死后PMN浸润,MPO活性增加,阿托伐他汀干预后可抑制PMN浸润和MPO活性,减少心肌梗死面积。

Abstract:Objective To investigate the effect of atorvastatin on the activity of MPO and the infarct size in acute myocardial infarction of rats. Methods AMI model was established by ligation of anterior descending branch of left coronary artery in male SD rats.Fifty-six SD rats were randomly divided into three groups; therapeutic, control and sham operation groups.The therapeutic group was pretreated with atorvastatin 10 mg·kg-1·d-1,the control group and sham operation group were pretreated with saline. Myocardial infarct size was assessed by TTC staining,and PMN were studied by HE staining and the number of the infiltrating PMN was measured by light microscopy.The MPO activity of the serum and the myocardium was measured by colorimetric method. Results The number of PMN increased significantly in the control group and therapeutic group compared with the sham-operation group(P<0.01),but was decreased significantly in the therapeutic group as compared with the control group (P<0.01). The activity of MPO was increased significantly in the control group and therapeutic group compared with the sham-operation group (P<0.01), but was decreased significantly in the therapeutic group as compared with control group (P<0.01). The infarct size in the therapeutic group was significantly smaller than that in the control group (P<0.01). Conclusion Acute myocardial infarction can increase the infiltration of PMN and the activity of MPO. Short-term (3days)atorvastatin treatment can significantly inhibit such changes,thus relieve AMI injury in rats and reduce myocardial infarct size, strongly suggesting that atorvastatin has protective effect on myocardium.
