摘    要:目的:研究桩蛋白(Paxillin)基因在人肺癌组织中表达,探讨它与肺癌患者病理特征、临床分期、淋巴结转移、预后的关系。方法:应用免疫组织化学S-P方法检测120例肺癌、80例癌旁无癌浸润的肺组织、53例正常肺组织的桩蛋白的基因表达。结果:肺癌、癌旁无癌浸润肺组织、正常肺组织桩蛋白的阳性表达率分别为39.1%(47/120),81.3%(65/80),83.3%(44/53),三者的阳性表达差异有高度显著性(P<0.05)。桩蛋白表达低下与肺癌P-TNM分期、病理分级、淋巴结转移有关(P<0.05),小细胞肺癌(SCLC)中桩蛋白的阳性表达率6.251%(1/16),显著低于非小细胞肺癌(NSCLC)桩蛋白的阳性表达率44.2%(46/104)(x~2=8.40,P<0.05)。结论:肺癌组织中Paxillin基因的表达低下与肺癌侵袭转移有关,可作为评价肺癌浸润转移的生物学标志之一。

Abstract:Objective: To detect the gene expression of paxillin in lung cancer tissues and to investigate the relationship between gene expressions of Paxillin and patient's P-TNM stages, pathological grades and lymph metastasis. Method: S-P immunohistochemical staining was applied to detect the gene expressions of Paxillin in tumor tissues from 120 eases of human primary lung cancer, normal adiacent lung tissues from another 80 cases and 53 cases of normal lung tissues. Results: The positive staining rate of Paxillin in primary lesion, adjacent lung tissues without carcinoma and normal lung tissues was 39.1% (47/120), 81.3% (65/80), 83.3% (44/53), respectively. The difference was highly significant (P<0.05). The positive expression rate of Paxillin protein of SCLC(6.25%, 1/16) were significantly lower than that of NSCLC (44.2%, 46/104) (x~2=8.40, P<0.05), and the low expression of paxillin of lung cancers was associated with histological grades, clinicopathological stages, and lymph node invading (P<0.05). Conclusion: Lower level of paxillin gene expression may cause lung cancer cells more aggressive and motile. It can be used as valuable biomarkers to evaluate lung cancer metastasis.
