电针“足三里”穴对大鼠T淋巴细胞亚群的影响

摘    要:本试验选用健康大鼠 ,随机分为正常对照组、“足三里”组、非经非穴组、免疫抑制组、免疫抑制 +“足三里”组。应用流式细胞仪技术 ,通过微量全血直接免疫荧光染色法测定各组大鼠的外周血T淋巴细胞亚群。实验结果如下 :电针“足三里”组大鼠T4细胞百分率明显高于正常对照组 (P <0 0 1 ) ,T8细胞百分率与正常对照组相比无统计学差异 (P >0 0 5 )。电针非经非穴组 ,各项指标与正常对照组相比无统计学差异 (P >0 0 5 )。且“足三里”组T4细胞百分率明显高于非经非穴组 (P <0 0 1 )。免疫抑制组T4细胞百分率明显低于正常对照组 (P <0 0 5 ) ,T8细胞百分率无明显变化 (P >0 0 5 )。免疫抑制 +“足三里”组大鼠T4细胞百分率与免疫抑制组相比有明显改善和提高 (P <0 0 1 )。结果显示 ,电针“足三里”穴可明显提高正常大鼠与免疫抑制大鼠的淋巴细胞亚群。而电针非经非穴点则无此作用。

Influence of Electroacupuncture at "Zusanli" on T-Lymphocyte Subgroups of Peripheral Blood in Rats

Abstract:Objective:To observe the therapeutic effect of electroacupuncture (EA) on T lymphocyte subgroups of peripheral blood in the rat.Methods: Forty Sprague Dawley rats were divided into five groups: control group, “Zusanli”(ST 36) group, immunosuppressive model group, none point group and immunosuppressive model+ EA group by blocked randomization. Immunosuppressive model was established by intraperitoneal injection of cyclophosphamide 100 mg/kg (twice). Bilateral “Zusanli” (ST 36) were punctured and stimulated with frequency 2 Hz and 3 V for 30 min, once daily, continuously for 7 days. Rats of the immunosuppressive and control group were only fixed to an animal stand for 30 min at the same hours every day. Blood samples were collected on the 7th day after intraperitoneal injection of pentobarbital sodium (1%, 30 mg/kg). The T lymphocyte subgroups of peripheral blood (including CD + 4 and CD + 8) were measured by direct microvolume whole blood immunoflurescent staining technique with flow cytometer(FCM) to evaluate the immune function of rats from different aspects. Results: ① The value of CD + 4 in EA group was significantly higher than that of control group(P<0.01),and the values of CD + 8 between the two groups had no statistical difference, indicating that EA had a markedly regulative effect on the immune function.② The value of CD + 4 in immunosuppressive model group was obviously lower than that of control group(P<0.01)and the values of CD + 8 between the two groups had not any statistical difference (P>0.05),indicating that cyclophosphamidum(CY)restrained the T lymphocyte subgroups of peripheral blood.③ The value of CD + 4 in the immunosuppressive model + EA group was significantly higher than that of immunosuppressive model group(P<0.01).④ The data of the none point group were not different from those of control group(P>0.05). Conclusion: EA on “Zusanli” point can improve cellular immune induced by T lymphocytes, but EA on none point has no such function.
