摘    要:目的:探讨三苯氧胺(tamoxifen,TAM)和阿霉素(adriamycin,ADM)联合应用对乳腺癌细胞MCF—7(ER )生长和凋亡的影响。方法:分别或联合应用不同浓度的TAM、ADM作用于MCF—7细胞,应用四甲基偶氮唑蓝(MTT)比色法分析细胞生长抑制作用,应用流式细胞技术测定细胞周期分布和凋亡率。结果:TAM与ADM均可抑制MCF—7细胞生长,诱导其凋亡。TAM可造成MCF—7细胞G0/G1期阻滞,且呈时间、剂量依赖性;ADM可造成G2/M期细胞增高;两者联用时MCF—7细胞的生长抑制率、凋亡率无明显增加,G0/G1期细胞增高。结论:TAM与ADM可抑制MCF—7细胞生长,诱导其凋亡,两者联合应用不能增强作用效果,无协同作用。

Abstract:Objective:To study the effect of tamoxifen and adriamycin on breast cancer cell line MCF-7. Methods:Estrogen receptor (ER)-positive MCF-7 human breast cancer cell line was treated by tamoxifen or adriamycin along , or in combination in vitro. Cell proliferation was evaluated by MTT assay. Distribution of cell cycle and rate of apoptosis were determined by flow cytometry. Results:Both tamoxifen and adriamycin can inhibit MCF-7 cells proliferation, and induce its apoptosis. The cells were arrested in G0/G1 phase by tamoxifen and G2/M phase by adriamycin along. The inhibition of cell proliferation and the change of cellcycle kinetics induced by tamoxifen were dependent on the does and time. When tamoxifen combined with adriamycin, the inhibitory effect on proliferation and apoptosis were not enhanced, but G0/G1 phase cells were raised. Conclusion: Both tamoxifen and adriamycin can inhibit MCF-7 cells proliferation, and induce its apoptosis, but therapy effect can not be improved when two drugs are combined. There is no synergism between them.
