摘    要:目的优选分离纯化骨碎补总黄酮的最佳大孔树脂,建立质量和收率稳定的骨碎补总黄酮制备工艺。方法以骨碎补总黄酮的含量和收率为指标,从大孔树脂的种类和用量、药液的吸附流速、洗脱用乙醇的浓度和流速、洗脱液收集点、洗脱乙醇用量等七方面设计优选骨碎补总黄酮的制备工艺。结果HPD-D大孔树脂分离效果最好,其最佳工艺为煎煮液过滤离心后,以吸附流速10mL/min.100g-1树脂进行吸附,用70%乙醇、4mL/min.100g-1树脂的洗脱流速进行洗脱效果最佳。结论HPD-D大孔树脂分离纯化骨碎补总黄酮制备工艺可行,收率和质量稳定,能够满足工业化生产需求。

Abstract:Objective To optimize the best macroporous resin in separation and purification of total flavonoids from Rhizoma Drynariae and establish preparation technique of Rhizoma Drynariae of stable quality and yield.Methods The content and the yield of total flavonoids of Rhizoma Drynariae were taken as the indexes.The preparation technique of total flavonoids of Rhizoma Drynariae was optimized in several aspects of the design,named the variety of macroporous resin,the dose of it ; absorption velocity of medical solution,concentration of alcohol in elution, the velocity of elution, collection of eluant and dose of alcohol in elution. Results HPD - D macroporous resin displayed the best separation. The optimum technique of preparation was designed as absorption at 10 mL/min/100 g resin after centrifugation of decoction ,and elution at 4 mL/min/ 100 g resin with 70% alcohol. Conclusion It is feasible to use HPD - D macroporous resin in separation and purification of total flavonoids from Rhizoma Drynariae. This preparation technique provides stable yield and quality and meets the requirements of industrial oroduction.
