加味半夏白术天麻汤对椎基底动脉供血不足眩晕大鼠血管活性物质的影响

摘    要:目的 观察加味半夏白术天麻汤对椎基底动脉供血不足(vertebrobasilar insufficiency,VBI)眩晕大鼠血管活性物质的影响.方法 将SPF级SD大鼠随机分为正常组、模型组、阳性对照组(用盐酸氟桂利嗪1.53 mg/kg灌胃治疗)、实验组(用加味半夏白术天麻汤141.7 mg/kg灌胃治疗),每组10只;将切除的一侧横突间骨翼置入骨膜下椎动脉内侧,以复制VBI眩晕模型;连续给药14 d,采用酶联免疫吸附试验测定各组大鼠血浆内皮素-1(endothelin-1,ET-1)、血栓素B2(thromboxane B2,TXB2)、6-酮-前列腺素F1α(6-keto-prostaglandin F1 alpha,6-Keto-PGF1α)、环磷酸腺苷(cyclin adenosine monophosphate,cAMP)的含量.结果 与正常组比较,模型组大鼠血浆ET-1、TXB2、6-Keto-PGF1α、cAMP含量以及TXB2/6-Keto-PGF1α比值均显著升高(P〈0.01);与模型组比较,阳性对照组和实验组上述指标均显著降低(P〈0.05,或P〈0.01).结论 加味半夏白术天麻汤对VBI眩晕的治疗作用与降低血浆ET-1和cAMP含量,恢复TXB2/6-Keto-PGF1α比值有关.

Abstract:Objective To observe the effect of modified Pinellia ternata, Atractylodes macrocephala, and Gastrodia elata Decoction on the vasoactive substances in rats with vertigo caused by vertebrobasilar insuf- ficiency (VBI). Methods Forty specific pathogen-free Sprague-Dawley rats were randomly divided into normal group (n = 10), model group (n = 10), positive control group (n = 10), and experimental group (n = 10). In the model group, positive control group, and experimental group, the resected wing of transverse process at one side was placed on the inner side of vertebral artery under the periosteum to in- duce VBI and establish a rat model of vertigo. The positive control group was given flunarizine hydrochlo- ride (1.53 mg/kg) by gavage for 14 d; the experimental group was given modified Pinellia ternata, At- ractylodes rnacrocephala, and Gastrodia elata Decoction (i41. 7 mg/kg) by gavage for 14 d. Enzyme- linked immunosorbent assay was used for measuring plasma levels of endothelin-1 (ET-1), thromboxane B2 (TXB2), 6-Keto-prostaglandin F1 alpha (6-Keto-PGFlo), and cyclic adenosine monophosphate (cAMP). Results Compared with the normal group, the model group had significantly increased plasma levels of ET-1, TXB2, 6-Keto-PGFlo, and cAMP and TXB2/6-Keto-PGF~o ratio (P d0.01). Compared with the model group, the positive control group and experimental group showed significant decreases in ~'He above indices (P d0.05 or P d0.01). Conclusion The action mechanism of modified Pinellia ternata, Atrac- tylodes macrocephala, and Gastrodia elata Decoction in treating vertigo caused by VBI involves decreasing plasma ET-1 and cAMP levels and restoring TXB2/6-Keto-PGFlo ratio.
