大豆异黄酮和运动对去卵巢大鼠股骨骨密度的影响

摘    要:目的:探讨大豆异黄酮与中等强度运动联合作用对去卵巢大鼠骨密度的影响。方法:将50只12周龄雌性Wistar大鼠按体重随机分为假手术组(Sham,正常对照组)、去卵巢组(OVX)、去卵巢+大豆异黄酮组(OVX+IF)、去卵巢+运动组(OVX+EXE)及去卵巢+大豆异黄酮+运动组(OVX+EXE+IF),每组10只。补药组大鼠以50mg/kg体重大豆异黄酮灌胃,其余组以等量溶剂灌胃。各运动组大鼠进行跑台训练。实验期包括2周适应期和8周训练期。实验前后称重。实验结束后处死大鼠,测量其股骨骨密度。结果:实验后,去卵巢组大鼠体重显著高于假手术组(P<0·01),去卵巢+运动组和去卵巢+大豆异黄酮+运动组大鼠体重均显著低于去卵巢组(P<0·05)。去卵巢组大鼠股骨骨密度显著低于假手术组(P<0·01)。去卵巢+运动组和去卵巢+大豆异黄酮组大鼠股骨骨密度与去卵巢组相比显著增加(P<0·05,P<0·01)。去卵巢+大豆异黄酮+运动组大鼠的股骨骨密度非常显著高于去卵巢组(P<0·001)。结论:大豆异黄酮和运动联合对抑制去卵巢大鼠骨密度减少有一定协同作用。

Abstract:Objective This study was designed to determine whether combination of soybean isoflavone supplementation and moderate intensity exercise would exhibit synergistically preventive effects on bone loss in ovariectomized rats Methods Fifty 12wk-old female Wistar rats were assigned to five groups: (1) Sham operated (Sham); (2) ovariectomized (OVX); (3) OVX with supplementation of soybean isoflavone (OVX-IF); (4) OVX with exercise (OVX-EXE); and (5) OVX with supplementation of soybean isoflavone plus exercise (OVX-IF-EXE).All rats were fed with normal diet ad libitum. Daily soybean isoflavone dosage was 50 mg/kg body weight. The vehicle was given to Sham, OVX-Cont and OVX-EXE groups. The soybean isoflavone were orally administered using a stomach tube. Rats in exercise groups were trained on an uphill treadmill at 20 m/min, 1h/day and 5days/week. The experimental duration consisted of the adaptation periods of 2 weeks and treatment periods of 8 weeks. Results The results showed that the body weights in OVX group was higher than that in Sham(P<0.01), OVX-EXE and OVX-IF-EXE had lower body weights than that in OVX (P<0.05). The femoral BMD (g/cm~2; mean +/- SD) of OVX was significantly lowered as compared to that of Sham (P<0.001). The BMD of OVX- EXE and OVX- IF increased significantly than that of OVX rats (P<0.05 and P<0.01,respectively). Combined treatment with soybean isoflavone and exercise revealed more successfully regaining BMD in ovariectomized rats (P<0.001). Conclusion These results suggested that the beneficial effects of combined treatment with soybean isoflavone and exercise was more obvious than separate acting of soybean isoflavone or exercise for preventing ovariectomy -induced bone loss in femurs.
