滨蒿内酯对大鼠血清Th1/Th2细胞因子及IgE表达的影响

摘    要:目的:观察滨蒿内酯(Scoparone,20ml/kg,浓度为168mg/L)对变应性鼻炎(AR)大鼠模型血清Th1/Th2细胞因子及IgE表达的影响,以探讨其治疗机制。方法:40只雄性SD大鼠,随机分为4组:正常对照组(NC组)、AR组、滨蒿内酯组(Sco组)和地塞米松组(Dxm组),以卵清蛋白致敏和激发建立大鼠AR动物模型并给予相应治疗。观察AR大鼠行为学改变;组织病理学方法观察鼻黏膜改变;采用ELISA法检测血清IgE和细胞因子IFN-γ、IL-4及IL-5水平。结果:Sco组行为学得分明显低于AR组,差异有统计学意义(P〈0.01),与Dxm组和NC组比较差异无显著性(P〉0.05)。Sco组鼻黏膜炎性反应明显轻于AR组。Sco组血清IFN-γ水平明显高于AR组,而血清IgE、IL-4和IL-5水平明显低于AR组,差异有统计学意义(P〈0.01);Sco组血清IFN-γ、IgE、IL-4、IL-5水平与Dxm组和NC组比较差异无统计学意义(P〉0.05)。结论:滨蒿内酯可通过调节Th1/Th2细胞因子的表达,抑制IgE的产生而对AR产生治疗作用。

Abstract:Objective:To observe changes of serum Th1/Th2 cytokines and IgE of rats with experimental al- lergic rhinitis(AR) treating with Scoparone (20ml/kg, the concentration is 168 rag/L). Method:Forty male SD rats were randomly divided into 4 groups:group NC,group AR, group Sco,group Dxm. The rats were sensitized with OVA and were then treated with Scoparone, during the treatment, the behaviors were observed and the change in nasal mucosa were recorded. The level of serum eytokines IFN-γ,IL-4, IL-5 and IgE were determined by ELISA. Result:The behavior scores of group Sco were significantly lower than those of group AR, the difference was statis tically significant(P〈0.01). Comparing to group Dxm and group NC respectively, the difference was not statisti cally significant(P〈0.05). The nasal inflammation of group Sco was significantly less than that of group AR. The level of serum IFN-γ of group Sco was significantly higher than that of group AR, while the levels of serum IgE, IL-4,IL-5 of group Sco were significantly lower(P〈0.01 ). There were no significantly difference between group Sco and group NC, group Dxm, the difference was not statistically significant (P〈 0.05). Conclusion: Scoparone is highly effective in treating allergic rhinitis by regulating the expression of Th1/Th2 cytokines and IgE.
