地龙成分及含药血清对人正常肾小球系膜细胞增殖的影响

摘    要:目的研究地龙成分EFE及含药血清对体外培养的人正常肾小球系膜细胞(HMC)增殖的影响,并从血清药物化学角度探讨其药理药效和防治糖尿病肾病的可能作用机制。方法将常规培养的HMC分为地龙成分EFE直接给药和含药血清给药两种方式,分别设空白对照组、低、中、高剂量组和福辛普利组,各组分别于相应处理后24 h、48 h和72 h采用4-甲偶氮唑蓝(MTT)法检测细胞增殖。结果直接给药组中各药物组与空白对照组(直)比较,均不同程度的抑制了正常HMC的增殖,中剂量药物组(直)对HMC细胞的增殖抑制作用最为显著;含药血清组中各药物组与空白对照组(血)比较,也不同程度的抑制了正常HMC的增殖,高剂量药物组(血)对HMC细胞的增殖抑制作用最为显著,并且高剂量药物组(血)对细胞增殖的抑制率高于中剂量药物组(直)。结论地龙成分能够抑制HMC的增殖,其含药血清对正常HMC增殖的抑制作用优于地龙成分直接给药。

Abstract:Objective To obseve the effect of earthworm composition and drug-containing serum to cultured human mesangial cells(HMC) and explore the pharmacological efficacy and possible mechanism in preventing diabetic nephropathy.Method HMCs of conventional culture were divided into two ways of direct delivery and drug-containing serum,what was the normal control group,the earthworm composition low,medium and high dose groups and the fosinopril group.Respectively given to the appropriate treatment after 24 h,48 h and 72 h detect cell proliferation with MTT method.Result Compared with the blank control group,each treatment group inhibited the normal proliferation of HMCs with different degrees.In which,the medium-dose group(direct) of the HMCs cell proliferation was the most significant,the high-dose group(serum)of the HMCs cell proliferation was the most significant,the high-dose group(serum)of the HMCs cell proliferation was better than the medium-dose group(direct).Conclusion Earthworm composition can inhibit the proliferation of HMC,its drug-containing serum showed better inhibitory effect than that of earthworm composition directly.
