二烯丙基二硫对人胶质瘤U251细胞增殖及RORα表达的影响

摘    要:目的探讨二烯丙基二硫(diallyl disulfide,DADS)对人胶质瘤U251细胞增殖及维甲酸相关孤核受体α(RORα)蛋白表达的影响。方法采用MTT比色实验、群体倍增时间分析DADS对人胶质瘤U251细胞的生长抑制效应;Western blot技术分析RORα蛋白在DADS处理U251细胞前后的表达改变。结果DADS浓度从15、22.5、30、37.5到45mg/L处理U251细胞,其吸光值比对照组低,而且随浓度增加逐渐降低(P〈0.05);相反,随着浓度增加,DADS对细胞增殖的抑制率从32.88%、41.83%、49.50%、59.42%到68.53%逐渐增强(P〈0.05);未处理的U251细胞群体倍增时间为21.86h,而DADS处理后的细胞,细胞群体倍增时间延长至36.69h(P〈0.05);Western blot结果发现,DADS处理U251细胞24、48h后,RORα蛋白表达(0.69±0.13、1.37±0.25)较未处理组(0.22±0.16)明显上调(P〈0.05),且48h处理组高于24h处理组(P〈0.05)。结论DADS可抑制人胶质瘤U251细胞增殖,其抑制增殖作用可能与诱导RORα蛋白表达上调有关。

Effect on proliferation and ROR alpha expression of diallyl disulfide induced human glioma cell line U251

Abstract:Objective To investigate the arrest effect of diallyl disulfide (DADS) in the proliferation and retinoic acid-related orphan receptor-alpha (ROR alpha) expression of human glioma cell line U251. Methods MTT assay,cell average doubling time analysis were employed to confirm the DADS-induced cell growth inhibition in cell line U251; Western Blot was used to observed expression of ROR alpha protein in U251 cells by 30mg/L DADS. Results Apparently growth inhibition in U251 cell line treated with DADS could be seen and exhibited a dose-dependent model. Exposure of U251 cells to 15mg/L DADS for 96h only yielded a ratio of 32.88%,whereas exposure to 30mg/L DADS increased the ratio up to 49.50%. Average doubling time retarded from 21.86 hours in normal cultured U251 cells to DADS experimented U251 cells 36.69 hours (P 〈0.05). Western Blot showed that the ROR alpha expression was increased after U251 cells treated with 30mg/L DADS for 24h and 48h,the gray scale value ratio of ROR alpha /beta-actin were 0.69±0.13 and 1.37±0.25 respectively,compare with 0.22± 0.16 as a control (P〈0.05). RORα expression in cells treated by DADS for 48h was higher than that for 24h (P〈0.05). Conclusion Apparently growth inhibition could be seen. In glioblastoma cell line U251 treated with DADS,in which up-regulate ROR alpha protein may be involved.
