精索内动静脉高位结扎iNOS的表达与睾丸生精细胞增殖和凋亡的关系

摘    要:目的:探讨精索内动静脉高位结扎术后,诱导型一氧化氮合酶(inducible nitric oxide synthase.iNOS)的表达与生精细胞增殖和凋亡的关系。方法:免疫组化结合计算机图像分析系统检测iNOS阳性细胞光密度值;免疫组化方法检测并计算PCNA增殖指数(PCNA-Labeling index,PI);原位末端标记法(terminal deoxynucleotidyl transferase mediated dUTP nick-endlabeling method,TUNEL)检测生精细胞凋亡指数(apoptosis index,AI)。分析iNOS的表达与增殖、凋亡的相关性。结果:iNOS在术后3-15d表达增强,第7d呈强阳性表达。结扎组生精细胞PCNA增殖指数(PI)明显低于对照组(P〈0.01),凋亡细胞指数(AI)明显高于对照组(P〈0.01)。结论:精索内动静脉高位结扎,iNOS的表达与生精细胞PCNA增殖指数呈负相关,与生精细胞凋亡指数呈正相关。iNOS基因表达增强是生精细胞增殖减少和凋亡增加的重要原因。

Abstract:Objective: To investigate the relationships of iNOS expression with spermatogenic cell proliferation and apoptosis following high ligation of internal spermatic cord vessels. Methods: iNOS was detected by immunohistochemistry combined with computer-assisted image analysis system. PCNA-labeling index(PI) was determined and calculated by immunohistochemistry, while spermatogenic cell apoptosis index(AI) was determined by TUNEL(terminal deoxynucleotidyl transferase mediated dUTP nick-end labeling). The relationships of iNOS expression with spermatogenic cell proliferation and apoptosis were analyzed. Results: iNOS expression increased 3-15 days following the high ligation, the high-positive expression of iNOS was detected on the 7th day after operation. PCNA-index(PI) in the high ligation groups were much lower than the control groups(P<0.01), while apoptosis index(AI) were higher than the control groups (P<0.01). Conclusions: After high ligation of internal spermatic cord vessels, the iNOS expression has negative correlation with PI of spermatogenic cell, and has positive correlation with AI. The over-expression of iNOS gene is one of the major reasons for decreasing of spermatogenic cell proliferation and increasing of apoptosis.
