白细胞介素2激活的外周血和脐带血及骨髓单个核细胞培养上清对骨髓造血的影响

目的：探讨用白细胞介素2（IL-2）激性的外周血（APB）和脐带血（ACB）及骨髓（ABM）单个核细胞的培养上清，对骨髓造血的影响和作用的可能机制。方法：应用MTT法，在体外同时检测APB和ACB及ABM培养上清，对骨髓和外周血单个核细胞增殖能力的影响；同时用集落培养法，检测这3种上清对骨髓粒-单细胞集落和成纤维细胞集落形成能力的影响。结果：加入APB和ACB及ABM培养上清，对正常骨髓和外周血单个核细胞增殖能力，以及粒-单细胞集落和成纤维细胞集落形成无显著的影响。结论：应用IL-2对自体干细胞移植物进行体外净化时，培养上清对正常造血细胞和单个核细胞无显著影响。

Effect of the Culture Supernatams of Interleukin-2-activated Peripher al Blood, Bone Marrow and Cord Blood Mononuclear Cells on Hematopoiesis.

Objective:To investigate the effect of the culture supernatants of interleukin 2 activated peripheral blood (APB), cord blood (ACB) and bone marrow (ABM) mononuclear cells on hematopoiesis. Methods: We employed MTT and colony culture methods to detect the effects of the supernatants of APB, ABM and ACB on normal bone marrow and peripheral blood mononuc lear cells, CFU GM and CFU F. Results: The supernatants of APB, ACB and ABM had no statistically significant effect on the proliferation of normal bone marrow and peripheral blood mononuclear cells, CFU GM and CFU F. Conclusion: While using interleukin 2 to purge the auto stem cell grafts with the contaminated tumor cells, the culture supernatants do not significantly affect the normal CFU GM and CFU F.
