摘 要: | 目的研究超声激活血卟啉诱导S180肿瘤细胞凋亡的作用机制,并探讨声动力学处理对线粒体凋亡相关蛋白表达变化的影响。方法采用频率为1.75 MHz,声强2W/cm~2的高频聚焦超声结合血卟啉处理S180腹水型肿瘤细胞,流式细胞仪检测细胞凋亡率;激光共聚焦扫描显微镜观察线粒体内膜通透性的变化和细胞色素C的释放;Western blotting检测线粒体凋亡相关蛋白Bcl-2、Bax和Caspase-3的表达变化。结果超声结合血卟啉联合作用后,S180肿瘤细胞凋亡率显著提高,线粒体内膜通透性增强,细胞色素C释放,Bax、Caspase-3蛋白的表达明显升高,而Bcl-2表达降低。结论线粒体凋亡通路在超声激活血卟啉诱导S180肿瘤细胞凋亡中可能起重要作用。

Effects of Mitochondrial Apoptotic Pathway in Induced Apoptosis of Tumor Cells by Sonodynamic Therapy |

Abstract: | Objective To investigate the underlying mechanisms of S180 apoptotic cell induced by ultrasound-activated hematoporphyrin derivative (HpD),and further discuss the changed expression level of mitochontria apoptosis related proteins by sonodynamic therapy.Methods Treated with high frequency focused ultrasound of 1.75 MHz at 2 W/cm~2 and HpD,S180 cell's apoptosis rate,mitochondria inner membrane permeability and Cytochrome C release,expression level of Bcl-2,Bax and Caspase-3 were examined by flow cytometer,confocal microscopy and Western blotting,respectively.Results The apoptosis rate,mitochondria inner membrane permeability,Cytochrome C release and Bax and Caspase-3 expression were greately enhanced by the synergistic effect of ultrasound and HpD,while the protein Bcl-2 was low expressed.Conclusions The mitochontria apoptosis pathway may play an important role in S180 apoptotic cell induced by ultrasound-activated HpD. |
