缬沙坦对高糖刺激下正常人类系膜细胞增殖状况及环氧化酶-2表达的影响

摘    要:目的 :研究高糖对正常人类系膜细胞 (NHMC)增殖作用及环氧化酶 2 (COX2 )表达状况 ,并观察用血管紧张素Ⅱ受体阻滞剂缬沙坦干预实验后对系膜细胞增殖的抑制作用和COX2 表达的影响。方法 :将NHMC置高糖状态下进行培养 ,并加缬沙坦进行干预实验。用WST 1法测定NHMC增殖情况 ,并用Westernblot技术 ,分析NHMC的COX2 表达情况。结果 :高糖组NHMC增殖明显高于低糖组。两组间差异有显著性意义 (P <0 .0 5 )。加入缬沙坦 (1.2 5 μmol/L)后 ,无论高糖组还是低糖组NHMC增殖均受到抑制 ,且随着缬沙坦剂量增加细胞增殖抑制递增。各浓度加药与非加药组比较 ,差异有显著性意义 (均P <0 .0 5或P <0 .0 1)。高、低糖组NHMC均有COX2 表达 ,加入缬沙坦后 ,COX2 表达减弱 ,特别是高糖组更为明显。结论 :①高糖能刺激NHMC增殖 ,缬沙坦能抑制NHMC增殖 ,②缬沙坦可降低COX2 在NHMC中表达作用 ,在高糖状态下其作用尤为明显。因此推测缬沙坦对糖尿病肾病的保护作用可能是通过其非动力学作用而实现的

Abstract:Objective:To investigate the effects of high glucose and valsartan on proliferation and cycooxygenase 2(COX 2) expression in NHMC.Methods:NHMC was cultured in vitro at high glucose concentration with or without valsartan. Wst-1 was employeed to determinate the proliferation and westernblot analysis was conducted for COX 2 expressions.Results:The proliferation in NHMC in response to high glucose was significantly higher than that to low glucose. These effects could be inhibited by valsartan in a dose-dependent manner. Valsartan also decreased the COX 2 expression in NHMC at high or low glucose concentration, which was more significant in high glucose condition.Conclusion:1.High glucose stimulates the proliferation in NHMC ,which could be inhibited by valsartan. 2.Valsartan could reduce COX 2 expression in NHMC, especially at high glucose concentration. The renoprotective effects of valsartan on diabetic nephropathy may be partially attribute to a non- hemodymanic mechanisms.
