RhoC及PTEN在非小细胞肺癌中的表达及临床意义的探讨

摘    要:目的 观察Ras同源性原癌基因C(RhoC)及张力蛋白同源的磷酸酶基因(PTEN)在非小细胞肺癌(NSCLC)组织中的表达程度及其与临床病理学之间的关系,探索RhoC与PTEN在肺癌发生及演变过程中的作用机制及两者之间的关系,为非小细胞肺癌的发生机制提供理论基础,并为NSCLC的早期诊断提供新思路.方法 (1)试验分组:60例非小细胞肺癌标本及40例正常对照肺组织.(2)采用免疫组织化学方法检测RhoC及PTEN表达情况及其与临床病理学之间的关系. 结果 60例NSCLC组织中, RhoC蛋白阳性表达率分别为68.33%(41/60),显著高于正常肺组织,RhoC在NSCLC组织中阳性表达率与吸烟史、年龄及组织学类型无关,而与癌组织的分化程度、临床分期密切相关.PTEN在非小细胞肺癌组织中的阳性率为36.70%,显著低于非肺癌组织的阳性率80.00%(P〈0.01) PTEN的表达与肺癌患者的细胞分化程度、TNM分期、肿瘤直径相关.RhoC和PTEN两者表达强度之间呈显著负相关(P〈0.01).结论 (1)在非小细胞肺癌组织中RhoC的高表达与PTEN的低表达密切相关,可能参与了NSCLC的演进.(2)PTEN基因蛋白表达缺失在不同类型和不同分化程度的非小细胞肺癌发生、发展中存在重要意义,可作为判断肺癌生物学行为和患者预后的参考指标.

Discussion on the expression and clinical significance of RhoC and PTEN in NSCLC

Abstract:Objective The aim of this study is to investigate the expression of ras homologus oneogenes C,phosphatase and tensin homology deleted on chromosome ten and clinical and pathophysiologieal characteristics in non - small cell lung cancer(NSCLC). To determine the effect of RhoC and PTEN in the processing of angiogenesis. Offer new basis for early diagnosis of NSCLC. Methods The expression of Rho C and PTEN was detected in 60 lung caneer tissues and 40 normal tissues as control by immunohistochemical staining technique. Results Positive rate of RhoC was 68. 33% (41/60) respectively in lung cancer tissue, which was higher than that in benign pulmonary tissues. Its expression was closely related to the grade of cell differentiation and TNM stage. PTEN expression in NSCLC was 36. 7% significantly lower than non - cancerous lung tissues 80. 0% ( P 〈 0. 01 ). The expression of PTEN gene significantly correlated with grade of cell differentiation , TNM stage, tumor diameter. There was significant negative correlation between the expression degrees of RhoC and PTEN (P 〈 0. 01 ). Conclusion RhoC protein was over - expressed and PTEN was low in NSCLC. The expression of RhoC and PTEN will be valuable for diagnosing lung cancer and evaluating malignancy extent and offer a new basis for early diagnosis of lung cancers. PTEN gene deletions might play a main role of tumor oceurrence and progress in NSCLC.
