强肌健力方含药血清对脾虚大鼠胸腺细胞分泌细胞因子的影响

目的：研究强肌健力方含药血清对脾虚大鼠胸腺细胞分泌肿瘤坏死因子-α（TNF-α）、白细胞介素-2（IL-2）、白细胞介素石（IL-6）的刺激作用。方法：制备强肌健力方含药血清，体外分离并培养胸腺细胞，应用血清药理学方法，观察不同浓度强肌健力方含药血清对体外培养的脾虚大鼠胸腺细胞分泌TNF-α、IL-2、IL-6水平的变化。结果：与胎牛血清组、空白血清组比较，三种不同浓度强肌健力方含药血清均能明显促进胸腺细胞分泌TNF-α、IL．2、IL-6含量（P〈0．05—0．01），尤以20％含药血清效果更佳（P〈0．01）。结论：强肌健力方含药血清能够干预脾虚大鼠胸腺细胞因子的分泌，具有免疫调节的作用。强肌健力方20％含药血清可能为最佳血药浓度。

Effect of Rat Serum From Animals Treated with Qiangjijianli Fang on the Secretion of Relevant Cytokines by Thymic Cells From Rat Models of Spleen-Deficency in Vitro

Objective To study the effect of rat serum containing Qiangjijianli Fang on the secretion of TNF-α, IL-2 and IL-6 by thymic cells from rat models of spleen-deficency in vitro. Methods Rat serum containing Qiangjijianli Fang was prepared from rats sacrificed after gastric garage with Qiangjijianli Fang （dose： 23.4g/kg/d） for 5 days. Thymic cells were prepared from rat models of spleen-defieency sacrificed after 20 days＇ treatment with rhuburb decoction and cultured with different strengths of rat serum containing Qiangjijianli Fang （10% , 20% , 30% , each n = 5 ） for 72h. The supematant contents of TNF-α, IL-2 and IL-6 were measured with RIA. Cultures without Qiangjijianli Fang containing sertma were used as controls. Results The thymic cells culture supematant con- tents of TNF-ot, IL-2 and IL-6 in all the cultures with different strengths of Qiangjijianli Fang containing serum （ 10% , 20% , 30% ） were significantly higher than those in cultures without Qiangjijianli Fang containing serum （with or without fetal bovine serum ） , （P〈0.05 -0.01）. The best results were obtained with 20% strength （P〈0.01）. Conclusion Rat serum containing Qiangjijianli Fang could enhance the secretion of TNF-α, IL-2 and IL-6 by thymic cell culture from rat models of spleen-deficiency in vitro, espe- cially at the strength of 20%.
