布洛芬微乳的制备及其透皮吸收研究

摘    要: 目的 制备布洛芬微乳,考察布洛芬微乳对离体小鼠皮的透皮能力。方法 Zetapals多功能电位/粒度分析仪测定布洛芬微乳的粒度及其分布。用稳定性实验考察布洛芬微乳的稳定性。用改进的Franz扩散池研究布洛芬微乳的透皮速率。高效液相色谱法测定布洛芬的含量。结果 布洛芬微乳液滴的平均粒径均为57.9nm。布洛芬微乳具有很好的稳定性。布洛芬的平均透皮速率为(189.79±11.71)μg·cm-2·h-1,透皮吸收行为符合Fick’s第一定律。结论 布洛芬微乳有很强的透皮能力,有望成为布洛芬的新型透皮给药制剂。

Abstract:OBJECTIVE To prepare O/W microemulsion formulation containing ibuprofen and investigate its in vitro transdermal delivery ability.METHODS The microemulsions were prepared and characterized using Zetapals Zeta potential/ particle size analysis instrument.The permeation flux of ibuprofen was determined in vitro using Franz diffusion cell fitted with mouse skin.The content of ibuprofen was measured by HPLC.The stability tests were performed to evaluate the ibuprofen microemulsions.RESULTS The average droplet size of the microemulsion was about 57.9 nm.A high permeation rate of (189.79±11.71)μg·cm-2·h-1 was showed in vitro.The permeation of microemulsion accorded with the Fick's first diffusion law.The studied microemulsion system showed good stability.CONCLUSION These results indicated that the studied microemulsion system with high permeation rate may be a promising vehicle for the transdermal delivery of ibuprofen.
