大鼠变应性鼻炎模型中T-bet和GATA-3的表达

摘    要:目的:探讨在大鼠变应性鼻炎(AR)模型单核细胞中T-bet/GATA-3基因的动态表达。方法:20只SD大鼠,雌雄各半,随机分为实验组和对照组,每组各10只。实验组用卵蛋白致敏,以氢氧化铝溶胶和灭活的百日咳杆菌为佐剂,卵蛋白滴鼻建立大鼠AR模型;对照组以氢氧化铝溶胶和灭活的百日咳杆菌生理盐水皮下注射,生理盐水滴鼻。分别在致敏结束、首次激发10h、末次激发10h心脏采血,分离血单核细胞。用RT-PCR法测定单核细胞中T-bet/GATA-3mRNA的表达,对结果进行统计学分析。结果:致敏结束、首次激发10h、末次激发10h3个时间点实验组T-bet mRNA的相对表达量分别为0.404±0.187、1.676±0.708、0.503±0.514,GATA-3mRNA的相对表达量分别为0.434±0.147、0.600±0.480、1.029±0.690;对照组T-bet mRNA的相对表达量分别为0.487±0.212、0.486±0.148、0.495±0.103,GATA-3mRNA的相对表达量分别为0.596±0.249、0.474±0.101、0.550±0.119。实验组首次激发后10hT-bet m...
