二氢石蒜碱对异丙肾上腺素致大鼠心肌缺血的保护作用

摘    要:目的:观察二氢石蒜碱(dihydrolycorine,DL)对大鼠心肌缺血损伤的保护作用。方法:采用异丙肾上腺素(Isoproterenol,Iso)制作大鼠心肌缺血模型,检测DL对缺血损伤后大鼠血清肌酸激酶(CK)和乳酸脱氢酶(LDH)活性、心肌组织及线粒体中超氧化物歧化酶(SOD)和谷光甘肽过氧化物酶(GSH-Px)活性及MDA含量以及心肌线粒体ATP酶活性的影响。结果:试验组大鼠缺血损伤后血清肌酸激酶(CK)和乳酸脱氢酶(LDH)活性明显升高,心肌组织与线粒体中SOD与GSH-Px活性显著下降,MDA含量明显升高,心肌线粒体ATP酶活性明显降低,与对照组比较具有显著性差异(P<0.01);随药物剂量增加(10、20、40 mg/kg)其血清CK和LDH活性逐渐降低,心肌组织及线粒体中SOD与GSH-Px活性逐渐回升,MDA含量逐渐趋于正常,ATP酶活性逐渐增加,以DL 20及40 mg/kg组作用更显著(P<0.01或P<0.05)。结论:DL对Iso致大鼠心肌缺血损伤具有保护作用,其机制可能与DL减少心肌组织脂质过氧化、提高心肌细胞抗氧化损伤的能力、减轻心肌能量代谢障碍有关。

Abstract:Objective To investigate the protective effect of dihydrolycorine on myocardial ischemia injury induced by isoproterenol sc in rats.Methods The myocardial ischemia injury model was developed by 20 mg/kg of isoproterenol sc in rats.The activities of serum lactate dehdrogenase(LDH) and creatine kinase(CK),the enzyme activities of superoxide dismutase(SOD),Glutathione Peroxidase(GSH-Px) and levels of malondialdehyde(MDA) content of myocardium and myocardial mitochondria as well as the activity of ATPase of myocardial mitochondria were all measured. Results In the model group,the activities of CK and LDH in serum were increased,the content of MDA in heart and mitochondria increased whereas the enzyme activities of SOD and GSH-Px of myocardium and myocardial mitochondria decreased.The activity of myocardial mitochondria ATPase was also decreased significantly(P<0.05 or P<0.01 vs normal group).In rats treated with 10,20 and 40 mg/kg dihydrolycorine,LDH and CK release and MDA content were decreased,while the enzyme activities of SOD,GSH-Px and ATPase were increased in a dose-dependent manner(P<0.05 or P<0.01 vs model group).Conclusion The results show that dihydrolycorine decreases the release of CK and LDH in serum and the levels of MDA in myocardium and mitochondria,improves the activities of total SOD,GSH-Px in myocardium and mitochondria as well as the activities of the mitochondria ATPase,which suggests that dihydrolycorine could protect myocardium from ischemia induced by isoproterenol in rat.
