大鼠脂肪变性供肝减体肝移植术后的肝再生

摘    要:目的 探讨大鼠脂肪变性供肝减体肝移植术后的肝再生方式及相关机制。方法 采用 79%标准饲料、2 0 %猪油、1%胆固醇混合喂饲 ,同时以 5 0 %乙醇灌胃每日 1ml/10 0 g ,时间为 4周 ,诱导供肝脂肪变性形成 ,大鼠 60 %减体肝移植模型。观察和比较术后 1、3、7、14d时PCNA、Br dU免疫组织化学及新鲜分离的肝细胞的流式细胞术结果及肝再生率。结果 脂肪变性供肝减体肝移植术后 1、3、7d的肝再生率较正常明显减低 (P <0 .0 1) ;各时点的PCNA标记指数 (P <0 .0 1)和BrdU标记指数差异均有非常显著性 (P <0 .0 1) ;脂变供肝术后的肝细胞增殖指数 (PI)在 7d时最高 (2 6.3 1% ) ,而正常供肝在 3d时最高 (4 2 .0 1% )。结论 大鼠脂肪变性供肝减体肝移植术后肝再生的高峰时间滞后、周期延长。

Abstract:Objective To explore the liver regeneration and its mechanism after reduced-size liver transplantation with steatotic donor in rats.Methods Steatotic liver model was established by feeding rats a diet consisting of 79% standard diet,20% lard and 1% cholesterol for 4 weeks.Rrat 60% reduced-size orthotopic liver transplantation was performed to explore the histochemistry of PCNA and BrdU,flow cytometry of the freshly isolated hepatocytes,and liver regeneration rate at 1st,3rd,7th and 14th day after operation.Results The liver regeneration rate of steatotic donors was significantly reduced in comparison with the normal group (2.16%,22.19%,52.96% on the day 1,3,7 respectively vs 7.31%, 47.85%,72.35%,F=96.5,122.4,20.1,P<0.01).The mark index (MI) of PCNA on the day 1,3,7,14 in steatotic graft was significantly reduced (1.88,11.9,47.3,25.3 vs 14.9,54.3,33.6, 9.3, t= 23.7,11.17,4.22,7.03,P<0.01),and MI of BrdU in steatotic graft was also significantly reduced (1.73, 12.3,31.6,15.1 vs 9.23,31.03,16.4,5.08,t=9.49,12.37,6.09,10.81,P<0.01).The peak time of proliferation in steatotic donor was on day 7 while that in normal donor on day 3.Conclusion The liver regeneration after reduced-size liver transplantation with steatotic donors was damaged and the peak time of proliferation was delayed,but proliferative cycle was prolonged.
