摘    要:目的 通过研究不同月份延胡索组织结构的变化规律及其所含生物碱的积累规律,对其生物碱进行组织化学定位。方法 采集不同月份野生延胡索块茎,制作石蜡切片及冰冻切片,通过普通显微镜及荧光显微观察组织结构变化规律及生物碱在其中的分布情况。结果 延胡索块茎总体呈新旧替代性生长;生物碱在除形成层外的各个组织中均有分布,其中靠近木栓层内部的部分含量较高;淀粉粒丰富处荧光更强。结论 生物碱含量与淀粉粒的多少可能成正相关;增加延胡索的体表面积与淀粉粒的多少有利于提高生物碱的含量。

Tuber Tissue Structure of Wild Rhizoma Corydalis and Histochemical Localization of Alkaloid

Abstract:Objective To investigate the histochemical localization of alkaloid by observing the changes in the tissue structure of Rhizoma Corydalis in different months and the accumulation of alkaloid. Methods The tuber tissue of wild Rhizoma Corydalis in different months was collected to prepare paraffin and frozen sections. A microscope and a fluorescence microscope were used to observe the changes in tissue structure and the distribution of alkaloid. Results The tuber tissue of Rhizoma Corydalis showed alternative growth of old and new tissues. Alkaloid was distributed in all tissues except the cambium, with the highest content in the phellem layer. Strong fluorescence was observed in the parts with rich starch granules. Conclusion The content of alkaloid may be positively correlated with the number of starch granules, and increasing the surface area of Rhizoma Corydalis and the number of starch granules may help to increase the content of alkaloid.
