丝氨酸蛋白酶级联相关性斯氏按蚊血细胞分析

摘    要:目的 分析与丝氮酸蛋白酶级联相关性的斯氏按蚊血细胞。方法 采用姬氏染色法观察血细胞类型,并利用免疫细胞化学技术检测斯氏按蚊血细胞内前酚氧化酶和丝氨酸蛋白酶,对酶学定位阳性细胞类型进行初步鉴定。结果 经姬氏染色,斯氏按蚊血细胞主要为颗粒细胞,前酚氧化酶和丝氨酸蛋白酶主要分布于颗粒细胞和浆细胞内。结论 斯氏按蚊血细胞可结构性表达前酚氧化酶和丝氨酸蛋白酶,颗粒细胞和浆细胞可能为丝氨酸蛋白酶级联成分来源的主要细胞类型。

Abstract:Objective To identify hemocytes related with serine protease cascade in Anopheles stephensi. Methods Hemocyte types were observed by staining with Giemsa' stain method. Both pro-phenoloxidase and serine protease in the hemocytes were assayed with immunocytochemistry technique. Then primary identification was made for pro-phenoloxidase- and/or serine protease-positive cells. Results By Giemsa'stain, most of the hemocytes were consisted of granular cells. Serine protease and pro-phenoloxidase were mainly distributed within granular cell and plasmatocyte. Conclusion Pro-phenoloxidase and serine protease are consti-tutively expressed by hemocytes in Anopheles stephensi. Granular cells and plasmatocytes might be the main source of serine protease cascade. (For Fig. 1 - 5, to see back cover)
