摘    要:目的建立人血浆中丙泊酚的检测方法。方法采用RP—HPLC法测定人脑脊液中药物浓度,色谱柱为Lichrospher C18(4.6×250mm,5μm),流动相为甲醇-水(72:28),流速1mL·min^-1.荧光检测λex为276nm、λem为312nm。结果在0.02—10mg·L^-1范围内丙泊酚峰面积与内标峰面积之比与浓度线性关系良好,最低检测浓度为0.02μg·L^-1,平均方法回收率为90.55%-97.45%,日内、日间RSD均小于10%。结论本测定方法操作简便、灵敏度高、专一性强、结果准确,适合于丙泊酚的临床治疗药物监测及药代动力学研究。

Abstract:Objective To develop a simple, sensitive and rapid method for the determination of propofol in human plasma. Methods The concentration of propofol was determined by RP - HPLC method. The analysis involved a 250 mmx 4.6mm column packed with C18(5 um) ; the mobile phase consisted of methyl alcohol -water (72:28 ) ; The fluorescence detective wave - lengths were : λex = 276 nm,λem = 312 nm. Results A good linearity was obtained in the range of 0.02 - 10 mg·L^-1 , the average recovery of propofol was between 90.55 97.45% ; the relative stand deviations of within -day and between day were less than 10% ; the minimal detectable concentration in plasma was 0. 02 μg·L^-1. Conclusion The result demonstrated that the method was simple, sensitive, accurate and reliable for the determination of propofol in human plasma, it was suitable for the pharmacokinetics study and clinical monitoring of propofol.
