摘    要:目的用分步筛选的方法检测致病性气单胞菌,以简化实验步骤,减轻工作强度。方法初筛项目为氧化酶、0%NaCl胨水、6%NaCl胨水3项试验,复筛项目为肌醇、精氨酸双水解酶、靛基质、赖氨酸脱羧酶等4项试验,凡氧化酶、靛基质阳性;0%NaCl胨水生长;6%NaCl不生长;肌醇阴性;精氨酸双水解酶、赖氨酸脱羧酶不定的菌株为疑似气单胞菌,再进一步作系统生化鉴定试验,确定其种类。结果检测海产品、鱼类、禽畜肉等3类样品共68份,经初筛检出疑似气单胞菌60株,检出率为85.3%。其中6份样品同时检出2种以上疑似气单胞菌,进一步复筛筛去非气单胞菌19株,提高筛选率31.7%;系统生化试验后,确定致病性气单胞菌41株,符合率为95.2%。结论分步筛选检测是一种较实用的过筛分离方法。简化了实验步骤,缩短了检测时间,减轻了工作量,节约了人力、物力,基本上解决了分离难的问题,取得了较满意的效果。

Abstract:Objective To develop a simply, rapid and convenient method to inspect Aeromonas. Methods A method of two-step filtration test is used to inspect Aeromonas. The prime filtration tests are the three ones: Oxidase, 0%NaCl, 6%NaCl peptone solution. The refiltration tests are the four ones: m-inositol, arginine, indole and lysine. Those strains, the results of which are positive for the tests of Oxidase, Indole and growth in 0%NaCl, but negative for the tests of 6%NaCl and m-inositol, variable for the tests of arginine and lysine, are the suspicious ones. Then systemic biochemical tests are done to identify their species. Results Totally 68 various samples were tested including seafood, fish and raw meat. Among all samples, 60 suspicious strains were found after the prime filtration tests, and the ratio of samples being detected is 85.3%. Especially, above 2 kinds of suspicious strains were simultaneously detected from 6 samples. Then 19 strains of non-aermononas, 31.7% of the suspicious ones, were discarded after the refiltration tests. Finally, 41 pathogenetic strains were identified through the systemic biochemical test, the ratio of coincidence is 95.2%. Conclusion The method of two-step filtration test is a banausic one. Comparing to the ordinary method, it is easier, more rapid and more convenient, and it can also save much more manpower and materials. Above all, the problem that Aeromonas is hard to be isolated is settled.
