摘    要:目的 :研究利用基因工程包装细胞进行基因转导与药物相结合来治疗脑胶质瘤的可行性。方法 :利用基因转导技术及分子生物学方法构建了逆转录病毒载体 STK及基因工程包装细胞psi2 STK,并利用 psi2 STK细胞和丙氧鸟苷 ( GCV)结合对胶质瘤细胞进行了离体的基因治疗研究。结果 :实验组 C6Bp BAG+ psi2 STK由于有 psi2 STK细胞的存在 ,对 GCV的敏感性比对照组C6Bp BAG+ psi2 STK明显增强。结论 :通过体外实验证实了逆转录病毒转导 HSV- TK基因 ,在培养 C6胶质瘤细胞中 ,提高了其对 GCV的敏感性

Abstract:Objective: In vitro , the gene therapy of mice malignant glioma was studied by the combination of psi2STK cells and ganciclovir (GCV). Methods:Retroviral vector STK and packaging psi2STK cell was constructed by the molecular biological technique. In this study, the kinetic analysis of psi2STK cells with vamous GCV concentrations, and sensitive analysis of C6BpBAG cells to GCV in psi2STK were performed. Results:C6 glioma derived cell lines and psi2STK were cultured in different concentrations of GCV. Compared with uninfected cells, infected cells were more sensitive to GCV. Conclusion: The sensitivity of HSV TK infected C6 cells to GCV was increased. In same concentration of GCV, difference was remarkable ( P <0.01). The study confirms that the gene therapy of brain glioma is possible.
