摘    要:目的:探讨吲哚乙酸(IAA)联合辣根过氧化物酶(HRP)对BXPC-3细胞凋亡及自由基表达的影响.方法:台盼蓝(曲利本兰)排斥试验,研究IAA/HRP对BXPC-3细胞死亡率的影响;原位细胞凋亡检测法检测细胞凋亡;生化法检测细胞内SOD,MDA的含量;激光共聚焦显微镜检测细胞内的自由基含量.结果:与对照组相比,IAA/HRP组的细胞凋亡数增加,而且随着IAA浓度的增大,细胞凋亡数存在上升的趋势;SOD活性和MDA含量在IAA/HRP组中均明显升高;对照组的自由基含量比IAA/HRP组明显降低,且自由基的含量具有浓度和时间依赖性.结论:IAA联合HRP诱导自由基产生是诱发人胰腺癌BXPC-3细胞凋亡的可能机制之一.

Abstract:AIM: To explore how indole-3-acetic (IAA) and horseradish peroxidase (HRP) affect the apoptosis and free radicals expression of BXPC-3 cells. METHODS: Trypan blue exclusion test was used to determine the mortality of BXPC-3 cells treated by IAA/HRP and in situ cell apoptosis detection kit was used to detect the apoptosis of BXPC-3 cells. Biochemical methods were employed to measure the production of SOD and MDA in BXPC-3 cells and the levels of free-radical in BXPC-3 cells were measured by Laser confocal microscope. RESULTS: The number of apoptosis cells between control group and IAA/HRP group had distinct difference in a dose-dependent pattern and the production of the SOD and MDA in BXPC-3 cells had distinct difference between control group and IAA/HRP group. The production of free radicals in BXPC-3 cells had distinct difference after administration of IAA/HRP in a time-and dose-dependant manner. CONCLUSION: The free radicals induced by IAA and HRP may be involved in apoptosis of BXPC-3 cells.
