中期保存的兔角膜内皮细胞的形态学观察

摘    要:目的 通过对兔角膜内皮细胞形态结构的观察 ,了解中期保存对内皮细胞的影响。方法 前房内注入M K液 ,全眼球保存兔角膜。在保存 1、3、5、7d后 ,分别进行角膜内皮显微镜检查、角膜内皮细胞活性染色及电镜学检查。结果 随着保存时间的延长 ,角膜内皮细胞的活性不断降低。尤其在保存 7d后 ,角膜内皮细胞存活率及六边形细胞百分率明显下降。超微结构显示细胞膜连续性破坏 ,细胞器丢失 ,细胞死亡。结论 角膜中期保存液保存角膜的最佳时间为 1~ 5d。

Abstract:Objective To investigate the effects of mid-term storage of cornea by observing the configuration of endothelial cells in rabbit.Methods Rabbit corneas were stored in the irrigations of anterior chamber with M-K liquid.Enucleation was performed after 1 day,3 days and 5 days respectively.Endothelial cells were actively stained by trypan blue.Morphological study was performed by specular microscope and electron microscope.Results The activity of endothelial cells was constantly dropped during the period of storage.The endothelium survival rate and the proportion of hexagonal cells were obviously dropped especially after 7 days.The scanning electronic microscopy showed that the gaps among the endothelial cells were enlarged and the connection of the cells was broken following storage.Conclusion The optimal period of mid-term stored corneas is during 1 to 5 days.
