摘 要: | 目的 检测Bmi-1在胰腺癌组织中的表达,分析其与胰腺癌临床病理特征之间的关系.方法 实时荧光定量PCR法检测22例手术切除的新鲜胰腺癌组织及配对癌旁组织中Bmi-1 mRNA的表达;免疫组织化学法检测61例胰腺癌石蜡块标本中Bmi-1、p16蛋白的表达,并分析与临床病理指标的关系.结果 22例胰腺癌组织中Bmi-1 mRNA的相对表达量为0.314±0.040,与配对的癌旁组织0.143±0.056的表达量相差显著(P<0.01).61例胰腺癌组织中36例(59.0%)Rmi-1蛋白表达阳性,34例(55.7%)p16蛋白表达缺失.胰腺癌组织Bmi-1的阳性表达与淋巴结转移相关(P=0.025);p16的表达缺失与淋巴结的转移(P=0.020)及肿瘤分化程度(P=0.018)相关;Bmi-1蛋白表达与p16蛋白表达呈负相关(P=0.033).结论 Bmi-1在胰腺癌组织中表达增加,它可能通过抑制p16表达调控胰腺癌的恶性生物学行为.

Abstract: | Objective To investigate the expression and significance of Bmi-1 in pancreatic cancer.Methods The expression of Bmi-1 mRNA was detected in pancreatic cancer and matched adjacent normal tissues by using real-time PCR in 22 cases of human pancreatic carcinoma.Bmi-1 and p16 protein were determined in 61 cases of human pancreatic carcinoma with immunohistochemistry and their relationship with clinicopathologic parameters was analyzed.Results The expression of Bmi-1 mRNA was 0.314 ±0.040 in cancerous tissues, and 0.143 ±0.056 in adjacent normal tissues, and the difference was statistically significant (P <0.01 ).36 cases (59.0%) of PC were detected with positive expression of Bmi-1 protein, and 34 cases (55.7%) with no expression of p16 protein.The expression of Bmi-1 was significantly correlated with lymph node metastasis ( P = 0.020 ) and tumor differentiation ( P = 0.018 ); the expression of Bmi-1 was negatively correlated with the expression of p16 ( P = 0.033 ).Conclusions Expression of Bmi-1 was increased in pancreatic cancer, which may be involved in the malignant behavior of pancreatic cancer by inhibiting p16 expression. |
