银杏黄酮苷元对人脐静脉内皮细胞ECV304增殖的影响

摘    要:目的:体外观察银杏黄酮苷元(GA)对人脐静脉内皮细胞ECV304增殖作用的影响,初步探讨其对内皮细胞的保护作用。方法:分别设对照组、空白组和实验组;实验组设GA6.25、12.5、25、50、100、500 mg/L共6个浓度孵育ECV304 48 h,25 mg/LGA与ECV304共同孵育6、12、24及48 h共4个时间点,采用MTT比色法观察GA对人脐静脉内皮细胞ECV304增殖的影响。结果:6.25~50 mg/L GA与ECV304共同培养48 h,细胞形态基本正常,细胞增殖率均大于100%,与对照组(未加药)比较差异显著(P〈0.05),且各组细胞增殖率随药物浓度增加而增加,P〈0.05;GA25 mg/L和脐静脉内皮细胞ECV304共培养6~24 h,各组细胞增殖率随作用时间延长而增加,各组间比较差异显著(P〈0.05)。结论:银杏黄酮苷元促进人脐静脉内皮细胞ECV304增殖的作用具有时间和浓度效应,提示GA具有保护血管内皮细胞的作用。

Abstract:Objective:To investigate the effects of ginkgetin aglycone(GA) on the proliferation of human umbilical venous endothelial cells,ECV304,and so as to explore the feasibility of GA in protecting vessel endothelial cells.Methods: ECV304 were cultured with GA of different concentrations for different hours.Cell proliferation rates were determined with MTT assay.Results: ECV304 cultured with GA of 50~6.25 mg/L for 6 to 48 hours were normal morphologically.Cell proliferation rates of different drug-groups all exceeded 100%,which was different from that of control group significantly(P0.05).Proliferation rates of ECV304 cultured with GA of 25~6.25mg/L for 6 to 24 hours increased along with the increase of GA's concentration and action time to some extend.Differences among the groups were significant(P0.05).Conclusions: GA promotes proliferation of ECV304 in a dose and time dependant manner.GA may have some protective effect on vessel endothelial cells.
