摘 要: | 目的探讨细胞周期D1(Cyclin D1)、P27、细胞角蛋白19(CK19)、ALDH1A1在乳腺病变中的表达及其临床意义。方法采用免疫组化法检测79例乳腺组织标本(包括正常乳腺、乳腺增生、乳腺导管原位癌、乳腺浸润性导管癌)中Cyclin D1、P27、CK19、ALDH1A1的表达情况,分析其与乳腺癌干细胞增殖及乳腺病变组织分化程度之间的关系。结果正常乳腺、乳腺增生、乳腺导管原位癌、乳腺浸润性导管癌中Cyclin D1、ALDH1A1的表达逐渐上升,P27、CK19的表达逐渐下降;乳腺正常及良性增生组织中Cyclin D1、CK19、ALDH1A1的表达明显低于乳腺导管原位癌及浸润性导管癌(P<0.05);乳腺正常及良性增生组织中P27的表达明显高于导管原位癌及浸润性导管癌(P<0.05)。结论 Cyclin D1、P27相互作用调节乳腺癌干细胞的发生及发展过程,CK19、ALDH1A1可作为乳腺癌诊断及恶性程度评估的参考指标。

Expression and significance of CyclinD1,P27,CK19 and ALDH1A1 in the lesions mammary gland |

Abstract: | Objective To investigate the express and significance of CyclinD1,P27,CK19 and ALDH1A1 in the benign and malignant lesions of human mammary gland.Methods A total of 79 samples of human mammary gland tissues(including normal mammary gland,hyperplasia,ductal primary carcinoma and infiltration ductal carcinoma)were collected in this study.Immunohistochemical technique was used to detect the express the CyclinD1,P27,CK19 and ALDH1A1.The correlation of their expression with the differentiation of the mammary gland tissues with benign and malignant hyperplasia was analyzed.Results The positive expression of CyclinD1 and ALDH1A1 gradually increased while the expression of P27 and CK19 reduced step by step in normal mammary gland,hyperplasia,ductal primary carcinoma and infiltration ductal carcinoma tissues.The expression of CyclinD1 and ALDH1A1 were significantly higher in benign and hyperplasia tissues than those in cancer tissues(ductal primary carcinoma plus infiltration ductal carcinoma)(P<0.05);the expression of P27 and CK19 were significantly lower in breast cancer tissues than those in breast tissues with benign hyperplasia(P<0.05).Conclusion CyclinD1 and P27 have closed relationship in breast cancer,they control the breast cancer stem cell cyclins and determine the differentiation of the cancer stem cells.Combined detection of CK19 and ALDH1A1 proteins maybe helpful for the pathological diagnosis of lesions of mammary gland. |
