膀胱平滑肌细胞与聚羟基丁酯的生物相容性

摘    要:目的 :评价生物可降解材料聚羟基丁酯 (PHB)与兔膀胱平滑肌细胞的生物相容性 ,探索PHB作为支架材料构建泌尿系组织的可行性。方法 :将膀胱平滑肌细胞与PHB共同培养 ,利用相差显微镜观察细胞的粘附和生长情况 ,利用MTT法检测细胞的粘附与增殖 ,并利用扫描电镜检测细胞在材料上的空间生长情况。结果 :膀胱平滑肌细胞在PHB上粘附良好 ,粘附率为 83.6 % ,PHB对细胞的生长和增殖无影响。结论 :兔膀胱平滑肌细胞与PHB细胞相容性好 ,PHB作为构建泌尿系组织的支架材料具有可行性。

Abstract:Objective: To evaluate the biocompatibility of polyhydroxybutyrate (PHB) with bladder smooth muscle cells. Methods: Rabbit bladder smooth muscle cells were separated and cultured with PHB in vitro. Cells were examined by phase contrast microscopy and scanning electron microscope(SEM) to assess cell growth and morphology. Cell adhesion and cell proliferation were detected with MTT assay. Results: Bladder smooth muscle cells adhered well to PHB with the adherence rate 83.6%. PHB had no effects on the survival and proliferation of bladder muscle cells. No obvious morphological changes were observed by phase contrast microscopy and SEM. Conclusion: All these indicates that PHB has a good biocompatibility with bladder muscle cells and PHB has a potential to be used as scaffold in urinary tissue engineering.
