布渣叶药材HPLC指纹图谱研究

摘    要:目的:建立布渣叶药材的HPLC指纹图谱分析方法,为有效控制其质量提供可靠的依据.方法:采用Waters XbridgeTMC18柱(4.6 mm ×250 mm,5 μm),流动相甲醇-0.05%磷酸溶液线性梯度洗脱,流速0.8 mL· min-1,检测波长320 nm,柱温25℃.结果:确定20个共有色谱峰为特征峰,构成布渣叶药材HPLC指纹图谱,其中3个共有峰得到确认,分别为牡荆苷、异牡荆苷和水仙苷;19批布渣叶样品中有15批样品指纹图谱与对照指纹图谱相似度>0.90,其余样品相似度>0.75.结论:该色谱方法简单、准确、重复性好,可用于布渣叶药材的质量评价.

Abstract:Objective:To establish a method of fingerprint analysis on Microcos paniculata by HPLC for the quality control.Method: The chromatographic fingerprints were obtained by the Waters Xbridge C18 column(4.6 mm×250 mm,5 μm) with the gradient elution solvent system composed of methanol and 0.05% phosphoric acid.The flow rate was 0.8 mL·min-1;the wavelength was set at 320 nm and the column temperature was maintained at 25 ℃.Result: Twenty main common peaks were selected in the standard fingerprint and three peaks of them were identified as vitexin,isovitexin and narcissoside.There were fifteen samples fingerprints with good similarities with correlation coefficients higher than 0.90 compared with the standard fingerprint in nineteen different samples of M.paniculata,while others were higher than 0.75.Conclusion: The method with good reproducibility is simple and accurate,it can be applied for quality assessment for M.paniculata.
