鞣花酸片在家兔体内药代动力学研究

目的:建立鞣花酸片血药浓度的高效液相色谱测定方法,并进行该药物在家兔体内的药代动力学研究.方法:采用Agilent C18(4.6 mm×250 mm,5μm)色谱柱,以乙腈-0.1％磷酸水(23∶77)为流动相,流速0.8 mL· min-1,检测波长254nm,柱温30℃.结果:家兔血浆中,鞣花酸的线性范围为0.013～1.635 mg·L-1(r=0.998 9),方法回收率＞95％,最低检测限为0.010 mg·L-1,家兔口服给药后,鞣花酸的药时曲线出现双峰,在家兔体内存在肝肠循环.结论:实验建立了家兔血浆中鞣花酸含量的HPLC测定方法,专属性强,可用于该药在家兔体内的药动学研究.

Pharmacokinetics of Ellagic Acid Tablets in Rabbits

Objective:To develop an HPLC method for the determination of ellagic acid tablets in rabbits plasma and to study the pharmacokinetics of ellagic acid tablets in rabbits.Method: An Agilent C18(4.6 mm×250 mm,5 μm) column was adopted,the mobile phase was a mixture of acetonitrile and 0.1% phosphoric acid solution(23∶77) at the flow rate of 0.8 mL·min-1,the detection wavelength was set at 254 nm and the column temperature was controlled at 30 ℃.Result: The retention time of ellagic acid was observed at 8.5 min.The linear range of calibration curve was within drug concentrations of 0.013-1.635 mg·L-1(r=0.998 9).The quantitation limit was 0.010 mg·L-1.The recoveries of three levels were more than 95%.The plasma ellagic acid concentration-time curve could be evaluated by the two compartment model.Conclusion: The developed HPLC method for the pharmacokinetic study of ellagic acid tablets is precise.
