大孔树脂在两性霉素B原料药纯化工艺中的应用研究

摘    要:通过考察多种型号树脂对两性霉素B原料药中杂质的吸附效果,筛选出几种对两性霉素B原料药中杂质有选择吸附性的树脂,然后对这几种树脂的吸附条件进行研究,确定了30倍树脂和500 mm:60 mm柱径比的最佳吸附条件.最后,利用不同极性树脂对两性霉素B原料药中杂质的吸附效果的差异性,研究不同树脂混合的最佳比例,结果发现在树脂M-1:M-2为3.6:6.4时,两性霉素B原料药中总杂质和最大单一杂质含量下降最明显.

Abstract:Through observing the adsorption effect of the different types of resins on impurities in amphotericin B to sift several most effective resins which have the selective extracting impurities and then their favorable technological conditions for adsorption were studied.The experiment results showed that the best adsorption condition was that the mass of the resin was 30 times and the chromatography column diameter ratio was 500 mm:60 mm.Finally utilizing the adsorption effect of the difference of the different polarity resins on impurities in amphotericin B,the best mixed proportions of the different resins were studied,whose result showed that when the ratio of resin M-1 and M-2 was 3.6:6.4,the total impurities and the biggest single impurities in amphotericin B declined most obviously.
