补肾活血汤对慢性心力衰竭大鼠血清sFas、MMP-2的影响

目的观察补肾活血汤对慢性心力衰竭大鼠血清可溶性凋亡调节因子（sFas）、基质金属蛋白酶-2（MMP-2）的影响。方法将32周龄自发性高血压大鼠（SHR）60只随机分为五个治疗组,每组12只;另12只正常Wistar大鼠作为空白对照组。治疗组分别给予补肾活血汤高、中、低剂量和卡托普利灌胃,模型对照组与空白对照组给予同剂量的蒸馏水灌胃。给药8周后,采用酶联免疫吸附法（ELISA）检测每组大鼠血清sFas、MMP-2的水平。结果与模型对照组比较,补肾活血汤高、中、低剂量组和卡托普利组大鼠血清sFas和MMP-2水平下降,差异有统计学意义（P〈0.05）;与卡托普利组比较,补肾活血汤高剂量组大鼠血清sFas和MMP-2水平显著下降,差异有统计学意义（P〈0.05）。结论补肾活血汤可降低慢性心衰大鼠血清sFas和MMP-2水平,从而减少心肌细胞凋亡和抑制左室重构,防治心力衰竭。

Impacts of Bushen Huoxue Decoction on the Levels of Serum sFas and MMP-2 in the Rats with Chronic Heart Failure

Objective To observe the impacts of bushen huoxue decoction on the levels of serum sFas(soluble apoptosis regulator)and MMP-2(matrix metalloproteinase-2)in the rats with chronic heart failure.Methods 60 rats with spontaneous hypertension were randomly divided into 5 treatment groups,12 rats in each group.There were other 12 normal Wistar rats in a blank control group.The treatment groups were treated with the gastric infusion of high-dose,middle-dose and low-dose bushen huoxue decoction as well as captopril separately.In the model control and blank control group,the gastric infusion with the same dose distilled water was administered.In 8 weeks,the enzyme-linked immunosorbent assay(ELISA)was used to detect the levels of serum sFas and MMP-2 for the rats in each group.Results As compared with the model control group,the levels of serum sFas and MMP-2 were reduced for the rats in the high-dose,middle-dose and low-dose bushen huoxue decoction groups as well as the captopril groups,presenting the statistical significant differences(P<0.05).As compared with the captopril group,the levels of serum sFas and MMP-2 were reduced significantly for the rats in the high-dose bushen huoxue decoction group,indicating the statistical significant difference(P<0.05).Conclusions Bushen huoxue decoction reduces the levels of serum sFas and MMP-2 for the rats with chronic heart failure,thus decreases cardiomyocyte apoptosis and inhibits the left ventricular reconstruction so that chronic heart failure can be prevented and further treated.
