摘 要: | 目的探讨甘草减轻黄药子的肝毒性作用。方法采用四甲基偶氮唑盐法(MTT法),观察不同剂量组、不同培养时间的肝细胞存活率,比较黄药子与甘草含药血清对体外培养的肝细胞存活率影响;用比色法检测细胞培养上清液ALT、AST、乳酸脱氢酶(LDH),分析黄药子与甘草配伍后肝细胞毒性的差异。结果黄药子高、中、低剂量组均可引起肝细胞存活率下降,存活率随着培养时间的延长及给药剂量的增高而降低;黄药子与甘草配伍组(配伍组)肝细胞的存活率显著提高,与黄药子高、中、低剂量组比较有显著差异,甘草配伍组ALT、AST、LDH活性受到抑制,与黄药子组比较有显著差异。结论黄药子含药血清对肝细胞有毒性作用,与动物给药剂量及细胞培养时间有关;黄药子与甘草配伍后,对肝细胞的毒性显著降低,甘草具有保护肝细胞的作用。

Abstract: | Objective To investigate the effect of Glycyrrhiza uralensis on alleviating hepatic toxicity caused by rhizoma dioscoreae bulbiferae based on the theory of sweet can relieve toxicity.Methods The survival rate of hepatocytes in different dose groups and different culture time was observed by MTT method.The effect of rhizoma dioscoreae bulbiferae and Glycyrrhiza uralensis medicated serum on survival rate of hepatocytes cultured in vitro was analyzed,the levels of ALT,AST and LDH in the culture medium were detected with colorimetric method,and the difference of hepatotoxicity was analyzed after using the combination of rhizoma dioscoreae bulbiferae and Glycyrrhiza uralensis.Results The survival rate of hepatocytes was decreased respectively in rhizoma dioscoreae bulbiferae groups with different doses,and the survival rate was decreased with the prolonging of the time and the increasing of the drug dosage.Compared with rhizoma dioscoreae bulbiferae group,the survival rate of hepatocytes was increased significantly,and the activity of ALT,AST and LDH was inhibited respectively in the combination of rhizoma dioscoreae bulbiferae and Glycyrrhiza uralensis group.Conclusion Rhizoma dioscoreae bulbiferae medicated serum can induce hepatic toxicity,which is related to the drug dosage and the time of cells culture.Glycyrrhiza uralensis can alleviate hepatic toxicity caused by rhizoma dioscoreae bulbiferae,and Glycyrrhiza uralensis has protective effect on hepatocytes. |
