摘    要:目的:转染wt-p53基因对SHG44细胞恶性表型的影响。方法脂质体介导法,将wt-p53基因转导入SHG44人胶质瘤细胞,斑点霜交证实转染成功;观察阳性转染细胞在体外常规培养中生长速率,细胞培增时间;平板集落形成率,结果转染wt-p53的细胞生长速度明显减慢,倍增时间延长(P〈0.01);细胞集落形成数明显减少(P〈0.01),结论外源性野生型P53基因对SHG44细胞的恶性表型有抑制作用。

Abstract:AIM To study the effect of wild type p53 gene transfection on malignant phenotype of SHG 44 cell line. METHODS Wild type p53 gene were transfected into human glioma cell line SHG 44 cells with liposome. The growth rate, cell double time and colony formation rate of transfected cells were observed. RESULTS The growth rate of wild type p53 gene transfected cells declined obviously, the double time extended ( P <0.01) and the number of colony formation significantly decreased ( P <0.01). CONCLUSION Exogenous wild type p53 gene could inhibit the malignant phenotype of SHG 44 cell line.
