山药总皂苷提取工艺研究

目的：优化山药总皂苷提取工艺。方法：采用UV法和HPLC法测定山药总皂苷和薯蓣皂苷,以二1者的含量为指标,通过单因素循环试验和正交设计试验考察乙醇体积分数、溶剂倍量、提取时间和提取次数对提取过程的影响,优化提取工艺。结果：确定最佳提取工艺为30倍量60%的乙醇回流提取三次,每次2h,总皂苷的含量为2.174mg·g-1,薯蓣皂苷的含量为0.790mg·g-1。结论：优化后的提取工艺稳定、可行,适用于山药总皂苷的提取。

Study on the Extraction Process of Total Saponins from Rhizoma dioscoreae

Objective：To optimize extraction process of total saponins from Rhizoma dioscoreae.Methods：Content of total saponins and dioscin as observing index were analyzed by UV and HPLC.Concentration of ethanol、extraction time、ratio of material to liquid and the times of extraction were optimized by using the single factor test and orthogonal design test.Result：The experiment proved that optimized process was using 30 folds amount of 60% ethanol for three times and extracting 2 h each time.Under this condition,the content of total saponins and dioscin were 2.17 mg·g-1 and 0.790 mg·g-1,respectively.Conclusion：The optimized process was stable,feasible and suitable for the extraction of total saponins from Rhizoma dioscoreae.
