雌/雄激素比例变化对大鼠前列腺基质细胞增殖和凋亡的影响

摘    要:目的:探讨雌/雄激素比例变化对前列腺基质细胞增殖和凋亡及雌激素受体表达的影响.方法:体外培养大鼠前列腺基质细胞,加入不同比例雌激素和雄激素,采用MTT法检测细胞增殖,以流式细胞术检测细胞凋亡,以细胞免疫荧光法检测雌激素受体亚型(ERα,ERβ)的表达.结果:雌激素(雌二醇,E2)和雄激素(双氢睾酮,DHT)联合应用时,可以促进基质细胞增殖,高比例的雌/雄激素可使基质细胞增殖增加(P<0.05)及凋亡减弱(P<0.05),且可以上调ERα的表达(P<0.05).结论:雌/雄激素比例升高使雌激素的促增殖作用增强而促凋亡作用减弱,这种作用是通过上调ERα的表达实现的.

Abstract:Objective: To investigate the effects of different ratios of estrogen to androgen(E2/DHT) on the proliferation and apoptosis of the cultured prostatic stromal cells as well as the expression of estrogen receptors(ERs,ERα and ERβ).Methods: The proliferation of cultured prostatic stromal cells under the action of different ratios of E2/DHT was detected by MTT method,the apoptosis was determined by flow cytometry(FCM),and the expression of ERα and ERβ was detected by immunofluorescence.Results: The combination of E2 and DHT enhanced the proliferation of prostatic stromal cells,and the higher E2/DHT ratio significantly promoted the proliferation(P<0.05) and decreased apoptosis of prostatic stromal cells(P<0.05).Under the action of higher ratio of E2/DHT,the expression of ERα was apparently higher(P<0.05).Conclusion: The increasing ratio of E2 /DHT promotes cell proliferation and inhibits apoptosis of prostatic stromal cells in vitro,which may be achieved through up-regulating the expression of ERα.
