赤芍和白芍抗炎作用的UPLC-MS代谢组学初步研究

摘    要:目的:通过比较赤芍和白芍对角叉菜胶致大鼠足趾肿胀的影响及内源性代谢物分析,探讨赤芍和白芍抗炎作用的异同.方法:建立角叉菜胶致大鼠足趾肿胀动物模型,观察赤芍和白芍的抗炎作用.基于超高效液相色谱-电喷雾质谱(UPLC-MS)比较赤芍和白芍给药后大鼠血清代谢物指纹图谱,采用偏最小二乘判别分析法研究给药组与正常组之间的代谢物组差异,寻找可能的生物标识物.结果:赤芍和白芍提取物在0.5~1h对大鼠角叉菜胶致足趾肿胀均有较好的抑制作用,在2~3h仅赤芍提取物有较好的抑制作用.前列腺素F2a,前列腺素E3,白细胞三烯A,前列腺素E2;谷胱甘肽等内源性生物标识物对于表征赤芍、白芍的抗炎作用具有重要的作用,且发现赤芍和白芍均可影响机体内前列腺素F2a和前列腺素E3的水平,但赤芍和白芍给药组间有显著性差异,提示二者发挥抗炎作用机制有一定差异.结论:代谢组学初步研究结果与传统药理试验结果有相关性,为进一步深人探讨赤芍和白芍的作用机制异同奠定基础.
