摘    要:目的 :研究脑出血大鼠脑内IL 6表达的细胞定位及脑溢安颗粒 (简称脑溢安 )对IL 6表达的影响 ,探讨脑溢安治疗脑出血的作用机制。方法 :采用胶原酶注射建立大鼠脑出血模型 ,以免疫组织化学方法检测脑出血后大鼠脑内白介素 6 (interleukin 6 ,IL 6 )的蛋白表达及胶质细胞反应。结果 :脑出血后 6h~ 4d血肿区域可以检测到小胶质细胞活性 ,6h~ 7d血肿外周可以见到星形胶质细胞反应 ;脑出血后 6h血肿区域即有IL 6表达 ,于 12h达高峰 ,7d时消失。结论 :脑出血后大鼠脑内IL 6阳性细胞主要为血肿区域的神经元和激活的小胶质细胞 ,脑溢安可能通过维持IL 6的上增性表达在脑出血损伤中发挥神经保护和神经营养作用

Abstract:Objective To define the cellular localization of IL 6 and the effects of the traditional Chinese medicine complex nao yi an granule (NYA) on the intercellular expression of IL 6 in the experimental intracerebral hemorrhagic (ICH) brain of rats, so as to investigate the efficacy mechanisms of NYA on ICH. Methods ICH rat models were established by stereotaxically injecting 0.4 U type Ⅶ collagenase into the right globus pallidus (GP). The distribution and content of IL 6 protein expression together with the reaction of glial cells were investigated with the immunohistochemistry method. Results Microglial activation was detected from 6 h to 4 d after ICH in the hematoma region, and reactive astrocytes were found from 6 h to 7 d after ICH around the hematoma region; there was an up regulation of IL 6 in the hematoma at 6 h after ICH; it reached its peak at 12 h, and disappeared on 7 d. Conclusion The major sources of IL 6 positive protein were neurons and the activated microglia in the brain of ICH rats. Stabilizing the increasing expression of IL 6 may be one of the neuroprotective mechanisms of NYA.
