高效液相色谱法测定布洛芬囊泡的包封率

目的：建立布洛芬囊泡包封率测定的高效液相色谱（HPLC）法。方法：色谱柱：Waters C18（4.6 mm×150 mm,5μm）,流动相：甲醇：水：冰醋酸（80：20：1,v/v/v）;流速：1 mL·min^-1;柱温：37℃;检测波长：228 nm;进样量为15μL。采用超滤离心法测定布洛芬包封率。结果：在此色谱条件下布洛芬与辅料及溶剂峰均得到良好的分离,布洛芬在10～100μg·mL^-1范围内峰面积与浓度呈良好的线性关系（r=0.9998,n=6）,平均回收率为100.5%,日内及日间精密度均小于2.0%（n=5）。结论：此方法准确可靠、简单快速,可用于布洛芬囊泡包封率的测定。

HPLC determination of entrapment effciency of ibuprofen niosomes

Objective：To establish a HPLC method to determine the entrapment efficency of ibuprofen niosomes.Methods：Using methanol-water-glacial acetic acid（80：20：1）as mobile phase,the separation was performed on a Waters C18 column（4.6 mm×150 mm,5μm）at a flow rate of 1 mL·min^-1.The detection wavelength was 228 nm and the column tempretature was 37 ℃.Results：Regression analysis of peak area against ibuprofen concentration showed good straight line fit over the range examined 10～100μg.mL^-1（r =0.9998,n =6）.Both the intra-day RSD and inter-day RSD were less than 2%（n =5.） And the average recovery was 100.5%.Conclusion：This method proves to be simple,accurate,sensitive and applicable for determination of entrapment efficency of ibuprofen niosomes.
