摘    要:目的:建立布洛芬囊泡包封率测定的高效液相色谱(HPLC)法。方法:色谱柱:Waters C18(4.6 mm×150 mm,5μm),流动相:甲醇:水:冰醋酸(80:20:1,v/v/v);流速:1 mL·min^-1;柱温:37℃;检测波长:228 nm;进样量为15μL。采用超滤离心法测定布洛芬包封率。结果:在此色谱条件下布洛芬与辅料及溶剂峰均得到良好的分离,布洛芬在10~100μg·mL^-1范围内峰面积与浓度呈良好的线性关系(r=0.9998,n=6),平均回收率为100.5%,日内及日间精密度均小于2.0%(n=5)。结论:此方法准确可靠、简单快速,可用于布洛芬囊泡包封率的测定。

Abstract:Objective:To establish a HPLC method to determine the entrapment efficency of ibuprofen niosomes.Methods:Using methanol-water-glacial acetic acid(80:20:1)as mobile phase,the separation was performed on a Waters C18 column(4.6 mm×150 mm,5μm)at a flow rate of 1 mL·min^-1.The detection wavelength was 228 nm and the column tempretature was 37 ℃.Results:Regression analysis of peak area against ibuprofen concentration showed good straight line fit over the range examined 10~100μg.mL^-1(r =0.9998,n =6).Both the intra-day RSD and inter-day RSD were less than 2%(n =5.) And the average recovery was 100.5%.Conclusion:This method proves to be simple,accurate,sensitive and applicable for determination of entrapment efficency of ibuprofen niosomes.
