C_3H非自发性小鼠乳腺癌模型的建立

摘    要:目的建立C3H非自发性小鼠乳腺癌模型。方法采用肿瘤组织块接种法和肿瘤细胞悬液接种法,将C3H自发性乳腺癌进行同种移植及异种移植,观察肿瘤生长情况,并连续传代。切除肿块作病理切片,免疫组化法检测雌激素受体(ER)、孕激素受体(PR)。结果>1 mm3体积的瘤块或>1×106细胞悬液移植接种,在接种后6~9 d在局部形成瘤性结节,继之迅速增大,成瘤率为100%;<0.5 mm3的瘤块和<0.5×106个细胞接种的成瘤率分别为40%和50%。肿瘤模型连续传代18代,生长稳定,传代周期固定。病理学检查为浸润性导管癌,ER阳性、PR阳性。结论该法建立的肿瘤模型,接种成功率高,成瘤时间周期短,适用于大面积的抗肿瘤药物实验。

Abstract:Aim This study was to establish a transplanted breast model of non-spontaneous breast cancer in C_3H mice.Methods The breast cancer tissue from spontaneous breast cancer C_3H was cut into several fragments sized 0.5~2 mm~3 or prepared into separated cells suspension of 0.5×10~6~2×10~9 cells·L~(-1).Then the fragments or suspensions were implanted or injected subcutaneously into forelimb axillas of the animals.The time,frequency of carcinogenesis and other characteristics of the cancer bearing animals were examined.The tumor masses were removed,and pathologically examined using microscope.Then ER and PR proteins were detected using immunohistochemistry respectively.Results After 6~9 days using cancerous nodes were formed at the site where >1 mm~3 cancer fragments or 1×10~6 cancer cells were implanted,then the cancer grew rapidly.The frequency of carcinogenesis was 100%.For the implantation with <0.5 mm~3 fragment or <0.5×10~6 cells,the frequency of carcinogenesis was lower.The tumor model had been implanted and passed 18 generations.All of the specimens were infiltrating ductal carcinoma,and expressed ER and PR proteins.Conclusion The model of non-spontaneous breast cancer in C_3H has been established.This model has a high frequency and short cycle of carcinogenesis.The model is suitable for large scale study in the experiments of antineoplastic drugs.
