益气养阴活血法对糖尿病大鼠肝脏P450的影响及其与血糖血脂的关系

目的探讨益气养阴活血法对糖尿病大鼠肝脏P450的影响及与血糖血脂的关系。方法取大鼠55只,分为健康对照组,模型组,丹蛭降糖胶囊(DJC)高、低剂量组,吡格列酮组,除健康对照组外,其余动物给予高脂饲料喂养4周,按35 mg/kg,给大鼠腹腔注射链脲佐菌素(STZ)溶液1次,造模成功后,DJC高、低剂量组分别给予1.08 g·kg~(-1)·d~(-1)、0.54 g·kg~(-1)·d~(-1),吡格列酮组给予10 mg·kg~(-1)·d~(-1),模型组、健康对照组给予等量0.9%氯化钠注射溶液。均连续灌胃8周,留取肝脏,逆转录反应及实时荧光定量PCR检测大鼠肝脏组织β-actin、P450,取血测定血糖、血脂、糖化血红蛋白(HbA1c)。结果与正常组比较,模型组大鼠肝脏P450表达明显升高(P0.01),予DJC后显著抑制了P450的过高表达,与模型组比较,差异有统计学意义(P0.01)。大鼠造模后血糖、血脂(TG)、HbA1c明显升高(P0.01),予DJC后,高、低两组血糖、血脂(TG)、HbA1c显著下降(P0.01或P0.05),疗效与吡格列酮近似。P450与血糖、HbA1c相关分析显示,均呈显著正相关(P0.01);P450与血脂各项指标均无明显相关性。结论具有益气养阴活血作用的丹蛭降糖胶囊对糖尿病大鼠肝脏P450酶活性有显著的抑制作用,有良好的降低血糖和血脂的作用,同时肝脏P450含量与血糖具有一定的相关性。

Influence of Tonifying Qi,nourishing Yin and activating blood method on the cytochrome P450 in the liver and its relationship with glucose and lipid

[Absract] Objective To explore the influence of Tonifying Qi,nourishing Yin and activating blood method on the cytochrome P450 in the liver of diabetic rats and its relationship with blood glucose and blood lipid.Methods Fifty-five rats were randomly divided into 4 groups:normal control group,model group,high-and low-dose Danzhi Jiangtang Capsule groups,and pioglitazone group.Except the control group,all others groups were fed with high-fat diet for 4 weeks,then gave diabetic model rats intraperitoneal injection with streptozotocin(STZ)of 35 mg/kg once only.The rats of high-dose DJC and low-dose DJC group were treated with DJCof 1.08 g·kg -1 ·d -1 ,0.54 g·kg -1 ·d -1 respective-ly,pioglitazone group was treated with pioglitazone of 10 mg·kg -1 ·d -1 ,the normal control group and model group were given saline 5 ml·kg -1 ·d -1 .After modeling,continuously administrate drug by drenching for 8 weeks,then put them to death and get the liver to do the tests of p450 and β-actin by the Real-Time PCR.Meanwhile,we measured the glucose,lipid and HbA1 c.Results Compared with the normal group,the expression of p450 in the model group in-creased significantly(P <0.01 ),the overexpression of p450 was significantly inhibited by DJC.compared with model group,the difference has statistical significance(P <0.01).After the model established,the level of glucose,lipid and HbA1 c were significantly increased(P <0.01),while the level of glucose,lipid and HbA1 c of high-dose DJC and low-dose DJC group were dacreased significantly.The effect of high-dose DJC and low-dose DJC were similar to pioglitazone group(P <0.01 or P <0.05).The correlation analysis showed that there was a significant positive correlation among P450 and blood glucose,HbA1 c.There was no significant correlation between P450 and lipid(P <0.01).Conclusion Danzhi Jiangtang Capsule can inhibit the activity of p450 enzyme in the liver of diabetic rats and reduce the blood glu-cose and lipid.Meanwhile,the P450 of liver and glucose is correlated.
